自固化磷酸钙复合兔自体骨髓基质细胞和bFGF异位成骨的研究
Experimental Research of Heterotopic Osteogenesis of Calcium Phosphate Cement Combined with Autologus Bone Marrow Stromal Cells and Basic Fibroblast Growth Factor
【摘要】 目的探讨自固化磷酸钙(CPC)复合自体骨髓基质细胞(BMSC)和碱性成纤维细胞生长因子(bFGF)异位成骨的可行性及其成骨效应。方法取兔骨髓,分离骨髓基质细胞,选取松质骨粒型自骨化磷酸钙置于板孔内,A组复合BMSC(3ml)和bFGF(100滋g/ml);B组仅复合BMSC(3ml);C组仅为CPC;置于培养箱中培养24h后植入兔骶棘肌内。分别于术后3天、4周、8周、12周行双能X线骨密度测定仪测定骨粒密度,光镜组织病理检测及图像分析并以激光共聚焦显微镜进一步观察,12周时行扫描电镜观察。结果术后8周A、B组骨粒的密度明显比C组高,12周时A、B两组也有显著性差异,光镜检测并以激光共聚焦显微镜观察术后8周A、B组有大量成骨样细胞,12周时有大量的板层状骨形成;而C组仍有较多的空泡,仅有少量的骨样组织。术后4周时A、B、C组的成骨面积差异有显著性。术后12周扫描电镜观察到A组的表面空隙内有明显的骨样组织填充,同时有多量的钙颗粒的沉积;B组次之,C组最少。结论bFGF可促进BMSC的增殖,BMSC能与自固化磷酸钙相容生长,自固化磷酸钙复合BMSC和bFGF可明显诱导新骨的形成。
【Abstract】 Objective: To investigate the osteogenesis of calcium phosphate cement(CPC) combined with autologus bone marrow stromal cells(BMSC) and basic fibroblast growth factor(bFGF). Methods: BMSC were separated from bone marrow of rabbits. Three groups were divided as Group A: CPC combined with BMSC(3 ml) and bFGF(100 ?滋g/ml), Groups B:CPC with BMSC(3 ml), and Groups C: CPC. Each group was implanted into the sacrospinalis muscle of rabbits after cultured for 24 h. Dual-energy X absorptionmetry(DEXA), histological observation and analysis, laser scanning confocal microscope(LSCM) and scanning electron microscope(SEM) were selectively carried out at 3 days, 4, 8, 12 weeks after implantation. Results: The DEXA showed the density of each group had significant differences at weeks 12 after operation.A great amount of osteoblasts were observed under histological microscope and LSCM at weeks 8 and plenty of lamellar bone tissues founded at weeks 12 after operation. Bone tissues in the gap of surface and calcareous deposited were observed under SEM at week 12. Conclusion: bFGF may promote BMSC proliferation; CPC have good compatibility with BMSC.CPC combined with BMSC and bFGF can promote osteogenesis significantly.
