肾上腺髓质素基因微卫星多态性与2型糖尿病的关联研究
Relationship between Microsatellite DMA Polymorphism of Human Adrenomedullin Gene and Type 2 Diabetes Mellitus
【摘要】 目的:探讨南京地区汉族人群中肾上腺髓质素(AM)基因多态性与2型糖尿病是否存在相关性。方法:选择AM基因附近高杂合度微卫星DNA多态标志,应用聚合酶链反应及变性聚丙烯酰胺凝胶电泳技术,探查这一微卫星多态在正常对照组与2型糖尿病组中的基因频率分布。结果:该多态位点存在4种等位基因,CA重复序列分别重复11、13、14和19次,病例与对照群体分析结果表明,两组间等位基因频率总体分布差异无显著性(x2=0.472,P=0.925)。两组间基因型频率总体分布差异亦无显著性(x2=7.332,P=0.501)。结论:南京地区汉族人群中肾上腺髓质素基因多态性可能与2型糖尿病无关。
【Abstract】 To explore the relationship between microsatellite DNA polymorphism of human adrenomedullin(AM) gene and type 2 diabetes mellitus in the Chinese population of Nanjing. Methods: Among Chinese of the Han nationality in Nanjing, 101 patients with type 2 diabetes mellitus and 108 nondiabetic subjects were collected randomly. Polymorphism of microsatellite marker was detected by polymerase chain reaction polyacrylamide gel electrophoresis and silver staining, to investigate the frequency distributions of AM gene microsatellite (CA)n in patients with type 2 diabetes mellitus or nondiabetic subjects. Results: In the Han Chinese, four types of alleles with different CA-repeat numbers were 11, 13, 14 and 19 respectively. The prevalence of AM gene allele frequencies in patients with type 2 diabetes mellitus was not significantly different from that in control subjects(x2 =0. 472, P = 0. 925) . Conclusion: Polymorphism of microsatellite markers in AM gene may not be associated with type 2 diabetes in the Han Chinese.
