Rock1基因在实验性肾间质纤维化大鼠肾组织中的表达及其意义

【摘要】 目的:观察Rock1基因在实验性肾间质纤维化大鼠肾组织中的表达规律,探讨Rock1在介导肾间质损伤与纤维化中的作用和意义。方法:采用逆转录-聚合酶链式反应(RT-PCR),观察单侧输尿管梗阻(UUO)大鼠不同阶段肾组织中Rock1mRNA表达水平的改变,及其与肾小管间质损伤相关指标和平滑肌肌动蛋白(α-SMA)mRNA表达水平的相关性。结果:Rock1基因在肾间质纤维化发生早期即明显上调(F=18.45,P<0.001),第7天达高峰而后下降,至UUO模型后期(第21天)降至基础表达水平。α-SMA基因表达在第3天即明显上调(F=18.43,P<0.001),第14天达高峰后有所下降,至模型后期仍高于基础表达。Rock1基因表达较α-SMA提前达到高峰,二者呈显著正相关(r=0.617,P<0.05)。Rock1基因与UUO大鼠肾间质炎性浸润程度呈显著正相关(r=0.673,P<0.01),并且在病变进展阶段与间质纤维化指标呈显著的正相关(r=0.664,P<0.01)。结论:Rock1基因在肾间质纤维化大鼠肾组织中出现了明显表达异常,它可能通过介导炎性细胞浸润和诱导细胞表型改变而参与了肾间质纤维化的形成。

【Abstract】 Objective:To study the expression of Rock1 mRNA in kidneys fro m ex perimental rat model of renal interstitial fibrosis (RIF) and the possible role of Rock1 in the progression of renal interstitial injury. Methods:The expressio ns of Rock1 and smooth muscle alpha-actin(á-SMA)were examined by RT-PCR in the obstructed kidneys from unilateral ureteral obstructed (UUO) rats, and the correlation between them was analyzed. The correlation between the expression le vel of Rock1 and interstitial fibrosis markers was also studied. Results:The ex pression of Rock1 was significantly up-regulated even before the onset of RIF ( F=18.45,P< 0.001).The expression of Rock1 was significantly positively correlat ed with that of á-SMA(r=0.617,P< 0.05), the degree of inflammatory influx(r=0. 673,P< 0.01)and index of interstitial fibrosis(r=0.664,P< 0.01)during the prog ressive phase of RIF. Conclution:Rock1 may be involved in the mechanism of the progression of RIF, and its effect may result from the induction of inflammatory influx and myofibroblast transdifferentiation.
