二硫氰基甲烷对啤酒酵母的影响

【摘要】 用生化的方法研究二硫氰基甲烷对啤酒酵母 (Saccharomycescerevisiae)的影响。结果表明 :该药剂对啤酒酵母的EC50 为 0 38mg·L-1 ,且对啤酒酵母孢子的形态没有影响 ;2 5mg·L-1 二硫氰基甲烷处理活化的啤酒酵母 5min ,对啤酒酵母的呼吸抑制率为 72 % ,处理 6 0min时 ,呼吸抑制率随着药剂浓度的增加而增加 ;用 1mg·L-1 二硫氰基甲烷处理啤酒酵母线粒体 ,对呼吸控制速率 (RCR)有 2 7 6 7%的抑制 ,磷氧比 (P/O)比对照 (水 )低 9 3% ;0 76mg·L-1 二硫氰基甲烷对琥珀酸细胞色素C还原酶和NADH细胞色素还原酶活性几乎没有影响 ,但对NADH氧化酶活性抑制率达 32 0 8%。

【Abstract】 Effects of metheylene dithiocyanate to Saccharomyces cerevisiae were studied by method of biochemical techniques EC 50 of methylene dithiocyanate against beer yeast( S cerevisiae )is 0 38 mg·L -1 .This chemical had no effect on the morphology conidium at the range of 03 2 mg·L -1 ,but can decrease 72% of conidium respiration of beer yeast compared with CK after treated 5 minutes with the concentration of 2 5 mg·L -1 .The higher the concentration was,the more the respiration decreased after treated 60 minutes,although yeast reacted differently after treated by different concentration during 530 min.Respiration of control rate( RCR )and the ratio of P/O were 27 67% and 9 3% lower than CK(water),respectively,when the mitochondria of yeast was treated at the concentration 1 mg·L -1 .Methylene dithiocyanate inhibited 32 08% NADH oxidase activity,one of electron transport system from cell free extracts from beer yeast,but with hardly no effect on succinic cytochrome C reductase and NADH cytochrome C reductase.
