金钗石斛种子非共生萌发和种质保存

【摘要】 报道了用金钗石斛(DendrobiumnobileLindl)已开裂果实的种子经无菌处理,播种在改良的KC培养基上培养,研究结果表明:种子萌发成原球茎,原球茎分化成幼苗。同时对原球茎、幼苗和茎芽进行冷冻保存和低温贮藏。最终研究结果为:冷冻保存的原球茎其分化率为51%,幼苗和茎芽存活率分别是35%、43%。低温贮存的原球茎分化率为75%,幼苗和茎芽的存活率分别是95%、97%。

【Abstract】 The seeds of split fruit of Dendrobium nobile Lindl were used in bacteria-free treatment and culture on the improved KC culture medium.The research showed seed sprouted into protocorm,which differentiated into seedling.Besides,the protocorm,seedling and stem bud were preserved in a freezing condition and microthermal condition.The final study showed the differential rate of protocorm under freezing preservation was 51%,the survival rate of seedling and stem bud were 35% and 43% respectively;the differential rate of protocorm under microthermal preservation was 75%,the survival rate of seedling and stem bud were 95% and 97% respectively.
