cDNA-AFLP结合BSA研究马铃薯晚疫病菌小种特异无毒基因差异表达片段

【摘要】 通过对具有 6个无毒基因不同表型的马铃薯晚疫病菌构建的 4个混合池进行cDNA AFLP分析 ,得到与 6个与无毒基因相关的差异表达片段 (TDFs) 85条 ,其中与无毒基因Avr1相关的为 2 0条 ,Avr2 2 8条 ,Avr3 Avr10 Avr1116条 ,Avr4 2 1条。差异表达片段大小主要分布在 10 0bp和 30 0bp之间 ,最小片段为 6 0bp ,最大片段 4 40bp。差异表达片段的获得将有利我们下一步克隆全长无毒基因和对晚疫病垂直抗性机制的研究

【Abstract】 cDNA-AFLP analysis on 4 pools consisted of Phytophthora infestans isolates bearing the different genotypes of six avirulence genes with 256 Apo+2 and Taq+2 PCs,resulting in 20 candidate TDFs for Avr1,28 for Avr2,16 for Avr3-Avr10-Avr11 and 21 for Avr4,so in total 85 interesting TDFs.These TDFs mainly distributed from 100 bp to 300 bp,the minimum size being 60 bp and the maximum size being 440 bp.Obtainment of these TDFS will be beneficial for us to clone full-length avirulence genes and study the mechanism of vertical resistance.
