电镜观察大鼠重症急性胰腺炎早期的肝脏损害
DETECT HEPATIC DAMAGE DURING EXPERIMENTAL SAP IN RATS BY TEM
【摘要】 目的 :为了观察重症急性胰腺炎 (SAP)时的肝脏功能改变及肝脏超微结构的变化。方法 :将 30只大鼠随机分为 3h、6h及 12h三个实验组和三个对照组 ,每组 5只 ,以胆胰管逆行注射法复制大鼠SAP模型。对照组仅做剖腹探查术。制模成功后 ,于上述时间点抽取股静脉血测定AST、ALT。切取实验组大鼠部分肝脏组织行病理组织学检查。结果 :显示SAP时 ,随病程延长 ,血清中AST、ALT浓度逐渐升高 ,与对照组相比 ,差异有显著意义。透射电镜 (TEM )观察可见肝细胞超微结构的破坏。结论 :SAP时 ,伴有明显的肝功能障碍及肝细胞结构破坏
【Abstract】 Objective: In order to detect hepatic damage during experimental SAP in rats. Methods: 30 male wistar rats were randomly divided into experimental and control group. SAP was induced by the retrograde injection of 5% sodium taurocholate into the pancreatic duct in experimental group. In the control group, laparotomy was performed simply. Rats were sacrificed 3?6 and 12h after the operation. At each point, the blood samples of femoral vein were obtained. AST and ALT levels were measured in the samples. Hepatic tissues in experimental group were used for histologic research. Results: With the development of SAP, the concentrations of serum AST and ALT were increased significantly in the experimental group as compared with the control. TEM demonstrated the irregularity of the hepatocellular nucleus, the damage of the mitochondria, the pyknosis of the nucleus and the leakage of the organella. Conclusions: Hepatic damage and disfunction occurred in SAP rats.
