不同保存期全血制备洗涤红细胞的超微结构变化
Analysis of the microstructure of red blood cells in whole blood during storage for preparation of washing red blood cell
【摘要】 目的 探讨不同保存期的全血制备洗涤红细胞前后电镜下红细胞形态的变化。方法 采集 CPD-A抗凝全血。实验分 ~ 组 ,分别于 4℃保存 7、1 0、1 5、2 0、2 5、3 0 d。于制备前取全血 1滴 ,5 %戊二醛固定 ,经 2 0 0 0 r/ min离心 1 0min后 ,分出血浆 ,取样检测血红蛋白。余下的红细胞再加等量生理盐水 ,1 5 0 0 r/ min离心 5 min,连续 2次 ,取样为制备后测定组。结果 组 和组 制备前后红细胞成双面凹的圆盘结构 ,细胞均匀混悬。组 制备前红细胞形态正常 ,制备后少数红细胞出现聚集状、球形或边缘不整齐 ,并有棘形红细胞出现。组 、组 、组 于制备前血浆微红 ;制备后 ,球形红细胞、棘形红细胞、中间漏孔的红细胞增加。结论 制备洗涤红细胞的最佳时间应在 4℃保存 1 0 d内的全血 ,保存 1 5 d以后的全血制备洗涤红细胞形态发生异常变化 ,出现棘形红细胞 ,囊泡化后的红细胞易溶血 ,红细胞寿命缩短 ,影响洗涤红细胞的质量。
【Abstract】 Objective To study the changes of microstructure of red blood cells(RBCs) in whole blood during storage for preparing washing red blood cells. Methods The whole blood was collected from healthy volunteers by using CPD-A collection bag. The experiment was divided into 6 groups. The group Ⅰ was the fresh whole blood stored at 4℃ for 7 days. The group Ⅱ and Ⅲ were the whole blood stored at 4℃ for 10 and 15 days, respectively. The group Ⅳ,Ⅴ and Ⅵ were the whole blood stored at 4℃ for 20, 25 and 30 days, respectively. 1 drop of whole blood taken from the whole blood bag was fixed by 5% glutaraldehyde before preparing washing red blood cell. The plasma was separated from the whole blood by 2 000 r/min for 10 min. The concentration of hemoglobulin (Hb) in the plasma was assayed. The left RBCs were added to equal volume of 0.9% NS and centrifuged twice at 1 500 r/min for 5 min. Results The shape of RBCs in the group Ⅰ and Ⅱ were double-faced concave disc and equably suspended. In the group Ⅲ, the RBCs before preparation showed the normal shape, but few RBCs after preparation were agglutinated, spherical and ragged at edge. The thorn RBCs could be found. The plasma was reddish and the number of spherical cells, thorn RBCs and middle-ventage RBCs increased before preparation in the group Ⅳ,Ⅴ and Ⅵ.Conclusion The best time of preparing washing RBCs should be fresh whole blood before 10 days stored at 4℃. The morphology of washing RBCs prepared from the whole blood would be abnormal after storage of 15 days.
