肺癌中凝血酶受体-1的表达及意义

【摘要】 目的 分析凝血酶受体 (PAR 1)在肺癌组织及细胞中的表达情况 ,探讨PAR 1与人肺癌生物学行为的关系。方法 采用免疫组织化学、Westernblot及RT PCR方法检测不同类型肺癌组织及细胞系中PAR 1的表达情况。结果 免疫组化S P法检测PAR 1在 5 2例肺癌组织中表达情况分别为 :腺癌 14 / 2 0例、鳞癌 6 / 16例、大细胞癌 6 / 10例、小细胞癌中 4 / 6例。免疫组化阳性细胞多位于癌巢周边 ,1例侵及支气管软骨处的鳞癌癌巢及另 1例低分化腺癌脉管内的癌栓呈明显阳性 ,1例腺癌组织周围的肺泡上皮不典型增生灶呈明显阳性。Westernblot及RT PCR方法检测结果均显示PAR 1在人肺巨细胞癌细胞系高转移株 (PLA80 1D)中的高表达明显高于其低转移株 (PLA80 1C)。结论 PAR 1在各种组织类型的人肺癌组织中均有不同比例表达。PAR 1在人肺巨细胞癌细胞系高转移株 (PLA80 1D)中的表达明显高于其低转移株 (PLA80 1C)。PAR 1表达可能与肺癌的恶性表型及生物学行为有关

【Abstract】 Purpose To analyze expression of PAR-1 in carcinoma of the lung and lung giant cell carcinoma cell lines and to explore the correlation between PAR-1 and the biological property of human lung carcinoma. Methods Receptor expression on protein and mRNA level was evaluated by using immunohistochemistry, Western blot, and RT-PCR techniques. Results Out of 52 cases of lung carcinoma examined, PAR-1 expressed in 14/20 cases of adenocarcinoma, 6/16 cases of squamocell carcinoms, 6/10 cases of large cell carcinoma and 4/6 cases of small cell carcinoma. Stronger PAR-1 staining was usually detected at the periphery of the cancer nests. Stronger PAR-1 staining was also detected at the periphery of one invasive squamocell carcinoma , in the cancer embolus of adenocarcinoma and in the foci of atypical adenomatous hyperplasia nearby the tissue of another adenocarcinoma. Lung giant cell carcinoma lines PLA801D (highly metastasis cell line) expressed higher mRNA and protein level of PAR-1 than PLA801C (lowly metastasis cell line) by RT-PCR and Western blot. Conclusions These results suggest that PAR-1 expressed in some lung carcinoma specimens and cell lines. Lung giant cell carcinoma lines PLA801D (highly metastasis cell line) expressed higher mRNA and protein level of PAR-1 than PLA801C (lowly metastasis cell line). PAR-1 may contribute to the malignant phenotype and biological behavior.
