苯并(a)芘对热休克因子1的影响
Effects of Benzo(a)pyrene on Heat Shock Factor 1
【摘要】 [目的 ]研究苯并 (a)芘 (benzo(a)pyrene ,BaP)对热休克因子 1(heatshockfactor 1,HSF1)的影响。 [方法 ]离体原代培养猪主动脉内皮细胞 ,以不同浓度的BaP( 0、0 .1、0 .5、1、5、10 μmol/L)染毒 2 4h ,分别以Western blot法和凝胶阻滞电泳 (electrophoreticmobilityshiftassay ,EMSA)法检测各浓度组细胞HSF1表达及HSF1与热休克元件 (heatshockelement,HSE)结合水平的变化。 [结果 ]低、中浓度 ( 0 .1~ 1μmol/L)的BaP对HSF1总表达量基本无影响 ,胞质和胞核HSF1表达量与阴性对照组相比没有明显改变 (P >0 .0 5 ) ,但随BaP浓度的升高 ,HSF1有向胞核转移的趋势 ;而高浓度 ( 5、 10μmol/L)的BaP会导致胞浆和胞核HSF1均降低 (P <0 .0 5 )。低、中浓度 ( 0 .1~ 1μmol/L)BaP作用下 ,HSF1 HSE结合水平基本不变或略有上升 ,高浓度 ( 5、10 μmol/L)的BaP使HSF1 HSE结合水平明显降低。[结论 ]BaP作用时 ,内皮细胞HSF1表达水平与分布、HSF1 HSE结合能力的改变可能是BaP导致热休克蛋白 (heatshockprotein 70 ,HSP70 )表达降低原因之一 ;但BaP作用下 ,内皮细胞HSF1表达水平与分布、HSF1 HSE结合能力的改变与HSP70表达改变并不完全一致 ,提示研究HSF1与HSE结合之后的改变对进一步揭示BaP致HSP70的基因表达降低的
【Abstract】 To investigate effects of benzo(a)pyrene on heat shock factor(HSF1) change in endothelial cells. Porcine aortic endothelial cells were primarily cultured in vitro,treated with BaP at different concentrations (0,0.1,0.5,1,5,10μmol/L) for 24 hours,then expression of HSF1 and binding levels of HSF1 and heat shock element (HSE) were determined using Western-blot and EMSA, respectively. Total expression of HSF1 was not significantly different between low,medium-concentration groups(0.1~1 μmol/L) and control group,there were no significant change in HSF1 expression in cytoplasm and nuclei of endothelial cells exposed to BaP compared with control group (P>0.05),but the expression slightly decreased in cytoplasm and slightly increased in nuclei with increase in BaP concentration; both expression of HSF1 in cytoplasm and nuclei had significantly decrease in high concentration groups(5~10μmol/L). The binding level of HSF1-HSE did not change or slightly increased in low and medium-concentration groups(0.1~1 μmol/L),but significantly decreased in high concentration groups(5~10 μmol/L) compared with control group. [Conclusion] The expression level and distribution of HSF1 and its binding levels to HSE might be related to the mechanisms of alteration of HSP70 gene expression;simultaneously,level and distribution of HSF1 and its binding levels to HSE did not change as same as alteration of HSP70 in endothelial cells exposed to BaP. It suggests that the further studies of the changes in detail after the binding of HSF1-HSE under BaP-exposed condition are important to understand HSP70 gene expression.
