苯并(a)芘对猪主动脉内皮细胞钙依赖型钾通道的影响
Effects of Benzo(a)pyrene on Ca2+-dependent K+ Channel of Porcine Aortic Endothelial Cells
【摘要】 [目的]研究苯并 (a)芘[benzo(a)pyrene ,BaP]对血管内皮细胞钙依赖型钾通道 (Ca2 + _dependentK+ channel,KCa)的影响及其意义。[方法]原代培养猪主动脉血管内皮细胞 ,不同浓度的BaP(0、0.1、0.5、1、5、10μmol/L)染毒24h,以全细胞膜片钳技术检测各组细胞KCa 的变化情况。[结果]随BaP浓度的逐渐升高 ,内皮细胞膜KCa 开放程度扩大。在膜电位为 +40mV时 ,对照组和BaP染毒组 (0.1、0.5、1、5、10μmol/L)的外向电流分别为 (350.12±53.23)、(372.35±54.56)、(765.25±86.55)、(897.44±79.33)、(1245.65±89.75)、(1810.23±65) pA ,其中BaP(0.5、1、5、10μmol/L)染毒组的外向电流值与对照组相比 ,差异有显著性(P<0.05)。[结论]BaP升高血管内皮细胞内Ca2 +浓度 ,可能引起KCa 开放程度扩大。
【Abstract】 Objective To investigate the role of benzo(a)pyrene on depolarization-evoked Ca 2+ -dependent outward K + currents(K Ca ).[Methods]Porcine aortic endothelial cells were primarily cultured in vitro,treated with BaP(0,0.1,0.5,1,5or10μmol/L)for24h,then K Ca was detected using whole-cell attached patch-clamp technique. [ Results] Depolarization-evoked Ca 2+ -dependent outward K + currents enhanced along with rise of BaP-concentration.When membrane potential was at+40mV,Ca 2+ -dependent outˉward K + currents of the control cells and BaP exposed cells(0.1,0.5,1,5or10μmol/L)were350.12±53.23,372.35±54.56,765.25±86.55,897.44±79.33,1245.65±89.75and1810.23±65pA respectively,the Ca 2+ -dependent outward K + currents of BaP-exposed cells(0.5,1,5or10μmol/L)increased significantly compared with that of control cells(P<0.01).[Conclusion]It is suggested that BaP increased Ca 2+ in cytoplasm and might cause enhancement of depolarization-evoked Ca 2+ -dependent outward K + currents.
