【摘要】 [目的]研究蛋白激酶C(PKC)信号途径在苯并(a)芘 (BaP)影响热休克蛋白70(HSP70)基因表达中的作用。[方法]离体培养猪主动脉内皮细胞 ,不同浓度BaP(0、0.1、0.5、1、5、10μmol/L)染毒24h,并以12四酸佛波酯13酸盐 (PMA)和抑制剂Staurosporine于不同时间点进行干预 ,以Western<sub>blot法检测各组细胞HSP70表达的变化。 [结果]BaP抑制HSP70表达 ;PKC的激动剂 (PMA)可完全或部分消除BaP对HSP70的抑制作用 ,而这种作用可迅速被PKC的抑制剂(Staurosporine ,STP)所阻断。[结论]BaP可能通过抑制内皮细胞PKC活性而抑制HSP70的表达

【Abstract】 Objective To investigate the role of protein kinase C in the change of HSP70gene expression caused by benzo(a)pyrene.[Methods]Porcine aortic endothelial cells were isolated and cultured in vitro.In BaP group,the cells were treated with BaP(0,0.1,0.5,1,5or10μmol/L)for24h.PMA were added into the medium after BaP(1or10μmol/L)treated cells for20h in BaP+PMA group,then the cells were treated additionally for4h.In BaP+PMA+staurosprine group,cells were treated with BaP(1μmol/L)for24h,PMA was added as above but staurosprine was added15minutes before adding PMA.In control group,the cells were treated only with dimethyl sulfoxide(DMSO).Expression of HSP70was determined by Western-blot.[Results]BaP inhibited the expression of HSP70.PMA,the activator of protein kinase C(PKC),weakened the inhibiting effects of BaP on the HSP70expresˉsion,which could be interrupted quickly by staurosporine,an inhibitor of PKC.[Conclusion]BaP might restrain HSP70expression through inhibiting activity of PKC.
