莪术油注射液对大鼠损伤血管的防治作用及胶原表达的影响

【摘要】 目的 :研究不同剂量莪术油注射液对大鼠球囊损伤血管新生内膜形成及血管重构的作用以及Ⅰ、Ⅲ型胶原的变化。方法 :术后 1 4d,计算机图像分析球囊导管损伤大鼠左颈总动脉形态学及组织学的变化 ,苦味酸天狼猩红 偏振光显示法结合图像分析检测各组Ⅰ型及Ⅲ型胶原的变化。结果 :模型组及莪术油小剂量组Ⅰ、Ⅲ型胶原较假手术组明显增多、增粗、变亮、紊乱 ,莪术油大剂量组较模型组则明显减少、变细、变暗、排列较整齐 ,各组Ⅰ、Ⅲ型胶原的分布均以外膜为主。模型组及莪术油小剂量组较假手术组腔面积、外弹力板周长、内弹力板周长显著减少 (P <0 .0 1 ) ,Ⅰ、Ⅲ型胶原的面积显著增加 (P <0 .0 1 )。莪术油小剂量组与模型组比较 ,差异无统计学意义 (P >0 .0 5 )。莪术油大剂量组和模型组比较 ,最大内膜厚度显著减少 (P <0 .0 1 )、新生内膜面积及Ⅰ、Ⅲ型胶原面积显著减少 (P <0 .0 5 ) ,腔面积、外弹力板周长、内弹力板周长显著增加 (P <0 .0 1 ) ;与莪术油小剂量组比较 ,差异有统计学意义 ;与假手术组相比 ,差异无统计学意义 (P >0 .0 5 )。结论 :Ⅰ、Ⅲ型胶原排列的紊乱、含量的增加是再狭窄形成的重要机制。大剂量莪术油注射液可能通过影响Ⅰ、Ⅲ型胶原合成显著抑制球囊损伤大鼠颈动脉内膜增生和血管重构

【Abstract】 Objective:To investigate the changes of type Ⅰ,Ⅲ collagen and the effects of different dosage of Oleum Curcumae Aromaticae oil injection on injured rat arteries after balloon angioplasty. Method:The left common carotid artery in rats was denuded with a balloon catheter. The rats were killed at 14 days after injury. Cross-sections stained with picrosirius red from the injuried segments were processed for distribution, arrangememt, amount of type Ⅰ and type Ⅲ collagen study by polarizing microscope and stained with hematoxylin-eosin for histological and morphological study by computer-assisted picture analysis system. Result:Type Ⅰ, Ⅲ collagen of model group and low-dosage Oleum Curcumae Aromaticae oil group were found mainly in adventitia, more thicker, brighter and more disordered than those in sham-operation group. Compared with model group and low-dosage Oleum Curcumae Aromaticae oil group, type Ⅰ and type Ⅲ collagen of high-dosage Oleum Curcumae Aromaticae oil group were found mainly in adventitia, less, thinner, dimmer, more orderly. Lumen area,EEL perimeter,iEL perimeter of model group and low-dosage Oleum Curcumae Aromaticae oil group were decreased (P< 0.01)and the areas of type Ⅰ,Ⅲ collagen increased significantly(P< 0.01)than those of sham-operation group, while there were not difference in these two groups (P> 0.05).Compared with model group,maximal intimal thickness,neointima area of high-dosage Oleum Curcumae Aromaticae oil group were decreased significantly (P< 0.01), as well as the areas of typeⅠand type Ⅲ collagen (P< 0.05),while there were not difference with those of the sham-operation group (P> 0.05). Lumen area,EEL perimeter,iEL perimeter increased markedly (P< 0.01)up to that of the sham-operation group (P> 0.05).The difference was significant between high-dosage and low-dosage group. Conclusion:The accumulation and derangement of collagen typeⅠ and Ⅲ may be important in restenosis. High-dosage Oleum Curcumae Aromaticae oil injection inhibits neointimal formation and vascular remodeling after arterial injury by the mechanisms of decreasing collagen typesiand Ⅲ production and well-arranging collagen typeⅠ and Ⅲ arrangement.
