近距离小剂量放疗对血管平滑肌细胞纤黏连蛋白及弹性蛋白基因表达的影响
Effects of radiotherapy on the gene expession of fibronectin and elastin in the cultured rat vascular smooth muscle cells
【摘要】 目的 :探讨近距离小剂量放疗对血管平滑肌细胞 (VSMC)纤维黏连蛋白 (FN)和弹性蛋白基因表达的影响及其机制。方法 :以 7、14、2 8Gy60 Co射线辐射培养的大鼠主动脉VSMC弹性蛋白及FNmRNA表达的影响 ,无辐射为对照组。结果 :7Gy放疗组对VSMC弹性蛋白及FNmRNA表达与对照组比较差异无统计学意义 ,而 14、2 8Gy放疗组FNmRNA分别为0 .72± 0 .12和 0 .4 9± 0 .0 9,与对照组 (0 .95± 0 .0 7)比较差异有统计学意义 (P <0 .0 5 )。 14和 2 8Gy辐射后细胞弹性蛋白mRNA水平分别为1.93± 0 .2 5及 2 .16± 0 .2 2 ,与对照组 (1.0 7± 0 .11)比较差异有统计学意义 (P <0 .0 5 )。结论 :14、2 8Gy60 Co射线辐射可以抑制FNmRNA及加强弹性蛋白mRNA的表达。
【Abstract】 Objective:To study the effects of radiotherapy on the gene expession of fibronectin (FN) and elastin in the cultured rat vascular smooth muscle cells (VSMCs). Method: Rat VSMCs were cultured in DMEM containing 10% FBS and were radiated by 60Co at a doses of 0,7,14,28 Gy respectively. The mRNA level of fibronectin and elastin were studied by reveries-transcriptative competitive PCR(RT PCR) . Result:The elastin mRNA levels were increased from 1.07± 0.11 to 1.93± 0.25 and 2.16± 0.22 in VSMC with 14,28 Gy radiation respectively (P< 0.05). The FN mRNA levels were decreased from 0.95± 0.07 to 0.72± 0.12 and 0.49± 0.09 in VSMC with 14,28 Gy radiation respectively(P< 0.05). Conclusion:radiotheapy inhabits the gene expression of FN and increased the gene expression of elastin in VSMC.
