质粒缺口-连接酶链反应-酶联免疫吸附法检测沙眼衣原体

【摘要】 目的:建立高度灵敏特异的质粒缺口-连接酶链反应-酶联免疫吸附法(Gap-LCR-ELISA)检测沙眼衣原体(Chlamy-diatrachomatisCT)。方法:根据CT共有隐蔽性质粒设计并标记4条寡核苷酸探针,对6型CT标准株和鹦鹉热衣原体及其他常见泌尿生殖道病原菌行Gap-LCR-ELISA。结果:质粒Gap-LCR-ELISA可检测出6型CT,并与其他病原体无交叉反应,其最低检测限为2.5个原体(elementarybodiesEB)比PCR敏感10倍。结论:质粒Gap-LCR-ELISA对检测CT感染具有极高的灵敏度和特异度,适宜于我国进行大规模CT流行病学调查。

【Abstract】 Objective: To establish a high sensitive and specific method (plasmid Gap-LCR-ELISA) for the detecting of Chlamydia trachomatis (CT). Methods: Four oligonucleotide probes were designed and labeled for plasmid Gap-LCR-ELISA, which were derived from CT cryptic plasmid. Then these probes were used to detect six standard serotype strains of CT and other bacteria causing the infection of genitourinary tract in clinic. Results: Six standard serotype strains of CT were detected specifically by Plasmid Gap-LCR-ELISA with no cross-reaction to the other bacteria. By this way CT could be detected in the concentration of 2.5 elementary bodies, which was 10 times as sensitive as PCR. Conclusions: Plasmid Gap-LCR-ELISA is an effective method to detect the infection of CT, which may be quite suitable for large-scale screening tests in our country.
