【摘要】 目的　检测复发尖锐湿疣患者的人乳头瘤病毒 (HPV)基因型 ,以探讨尖锐湿疣复发的机理 ,并指导治疗。方法　以蛋白酶K消化法提取 81例复发尖锐湿疣组织临床样本DNA ,以各型HPVDNA通用引物行PCR扩增 ,将荧光偏振检测技术与模板指导的末端延伸反应 (TDI FP)结合 ,应用HPV 6B ,11,16 ,18各型特异探针杂交延伸反应对扩增产物进行HPV分型 ,并分别以含HPV 6B ,11,16 ,18L1质粒的阳性及不含待测靶基因的同源质粒的阴性标准品为参照。结果　 81例尖锐湿疣组织块中HPV检出率 10 0 % ,其中两型混合感染 2 9例 ,占 36 .6 % ,三型混合感染 7例 ,占 8.6 %。结论　本组 81例复发尖锐湿疣患者的HPV的多重感染率高达 4 5 % ,可能与该病的反复复发有一定相关性。更多还原

【Abstract】 Objective To study the recurrent mechanism of condyloma acuminatum.Methods The genotypes of human papillomavirus (HPV) in 81 cases with recurrent condyloma acuminatum (CA) were investigated.Common conservative primers were used to amplify HPV DNA in the samples from condyloma acuminatum patients by PCR.The PCR products were identified and genotyped by template directed dye-terminator incorporation of fluorometric-polarization (TDI-FP) in which type-specific probes(HPV6B,11,16 and 18)hybridization and special fluorogenic labeled ddNTP incorporation were performed.The fluorogenic ddNTP was directly added to the end of probe.The results were measured by fluorometric-polarization microplate reader.Results The positive rate of HPV was 100% in 81 CA cases.Double types of HPV infection were found in 29 cases,and triple types of HPV infection were found in 7 of recurrent CA patients.The rate of multiple types of HPV infection in recurrent CA patients was 45%.Conclusions The multiple types of HPV infection may be related with recurrence of CA.更多还原
