微小颗粒骨异位成骨过程中TGF-β1的表达

【摘要】 目的 通过观察转化生长因子 β1(TGF β1)在自体微小颗粒骨 (30 0~ 5 0 0 μm)异位成骨过程中的表达 ,探讨微小颗粒骨移植的成骨机制。方法 采用日本大耳白兔造股二头肌肌袋模型 ,分别植入自体微小颗粒骨及自体块骨。术后 1、3、5、7、11、14、2 1、2 8d取材 ,进行组织学、免疫组化染色及原位杂交。检测TGF β1及TGF β1mRNA表达并作图像分析。结果 ①术后 5d ,颗粒骨组开始有软骨生成 ,7d时达到高峰 ,2 1d后有骨吸收 ;块骨组则以骨吸收为主。②术后 1d见基质及血肿中TGF β1阳性染色 ,亦见骨细胞TGF β1阳性染色。 5~ 11d颗粒骨组见成骨细胞、软骨细胞、间充质细胞及骨细胞表达大量TGF β1。 14d以后TGF β1渐减少 ,2 1d以后渐平稳。颗粒组TGF β1表达高峰出现早、强度高、持续时间长。③颗粒骨组TGF β1mRNA阳性表达细胞出现早 ,高峰在术后 5~ 11d ,主要为软骨细胞、成骨细胞、骨细胞及间充质细胞。结论 颗粒骨异位成骨能力强于块骨 ,TGF β1表达高峰出现时间早、量大、持续时间长。TGF β1mRNA定位细胞广泛 ,信号明显强于各期块骨组。

【Abstract】 Objective To observe the expression of TGF-β 1 in micromorselized bone autograft ectopic osteogenesis and study the osteogenesis mechanism of micromorselized bone. Methods Japanese biger rabbits were used to make bag model of the biceps muscle of arm. Autograft micromorselized bone and bulk bone was then implanted to rabbits, respectively. The samples were harvested on the 1st, 3rd,5th,7th,11th,14th,21st,28th day postoperatively, and examined by histology, immunohistochemistry staining and in situ hybridization analysis. The expression of TGF and TGF-mRNA were assayed. Results ① In the area of morselized bone, cartilage formation was found on the 5th day after operation,and reached peak on the 7th day, then slowed down till the 21st day .While in the area of bulk bone, absorption was the main reaction. ② Osteocyte matrix and hematoma TGF-β 1 positive staining were found on 1st day after operation. Massive expression of TGF-β 1 in chondrocyte, osteoblast, mesenchymal cell and osteocyte TGF-β 1 was found between the 5th and 11th day, and began to decrease in the 14th day, and kept steady after the 21st day. The expression peak of TGF-β 1 appeared longer than that of bulk bone. ③ In the area of morselized bone TGF-β 1mRNA positively cell appeared earlier and the peak was between 5th and 11th day. Most of cells were chondrocyte osteoblast, osteocyte and mesenchymal cell. Conclusion The capacity of ectopic osteogenesis of morselized bone is better than that of bulk bone. TGF-β 1 expression peak appears earlier, and expresses more, and lasts longer compared with bulk bone. TGF-β 1 mRNA staining cell is more, and signal is stronger than those of bulk bone in the corresponding period.
