丹参注射液诱导骨髓基质细胞分化为神经元的研究

【摘要】 目的 :探讨丹参注射液体外诱导成年大鼠骨髓基质细胞分化为神经元样细胞 .方法 :用贴壁法分离纯化SD大鼠骨髓基质细胞 .培养传至第 5代 ,以 10 %丹参注射液诱导骨髓基质细胞分化 ,6h后相差显微镜观察形态变化及免疫细胞化学染色鉴定神经细胞特异性标志物NSE及GFAP .结果 :诱导 1h后部分骨髓基质细胞胞体收缩 ,有突起伸出 ;6h后突起增多形成网络结构 ,免疫细胞化学染色 ,NSE强阳性表达率为(43 3± 4 3) % ;而GFAP无阳性表达 .结论 :丹参可以诱导成年大鼠骨髓基质细胞在体外分化成为神经元样细胞 .

【Abstract】 Objective: To investigate the differentiation of adult rat’s bone marrow stromal cells (BMSCs) into neuron-like cells with salvia miltiorrhiza injection. Methods: The bone marrow stromal cells of SD rat were separated and purified by passage culture. The 5th passage of BMSCs was induced by 10% salvia miltiorrhiza injection, the differentiation was observed under phase contrast microscope and the cells were stained immunocytochemically with NSE and GFAP. Results: After an hour of induction, partly cytoplasm of BMSCs contracted with protruding process; the immunocytochemical staining was performed on cells after induction for 6 hours, the rate of NSE staining positive was (43.3±4.3)%, but negative in GFAP immunocytochemical staining. Conclusion: Salvia miltiorrhiza could induce adult rat BMSCs differentiate into neuron-like cells.
