SHV-28型β内酰胺酶的原核表达及其性质鉴定
Expression of SHV-28 β-lactamase gene in prokaryote cells and identification of SHV-28 β-lactamase protein
【摘要】 目的:获得SHV-28型β内酰胺酶蛋白,并研究其特性。方法:将SHV-28型β内酰胺酶基因连接到pET-28b载体上,重组质粒pET-28b/SHV-28转化大肠埃希菌BL21(DE3)。用酶抑制剂增强的纸片扩散法检测重组菌的表型,等电聚焦电泳测定其表达蛋白的等电点。结果:SHV-28型β内酰胺酶的等电点为7.6.为非超广谱β内酰胺酶。结论:SHV-28型β内酰胺酶蛋白表达正确,为进一步研究新基因型β内酰胺酶奠定了基础。
【Abstract】 Objective: To investigate the expression of SHV-28 β-lactamase gene and identify the properties of its protein. Methods:SHV-28 gene was ligated with pET-28b. Recombinant plasmid pET-28b/SHV-28 was transferred into E. coli BL21 (DE3) cells by transformation. The phenotype of this recombinant strain was identified by β-lactamase inhibitor potentiated disk diffusion test. The isoelectric point (pI) of the protein isolated from the recombinant strain was determined by isoelectric focus. Results:Result of inhibitor potentiated disk diffusion test indicated that SHV-28 was not an extended-spectrum β-lactamase, with pI of 7. 6. Conclusions:SHV-28 gene can be expressed in prokaryote cells, and this forms the basis for further study of new β-lactamases.
