大鼠睾丸Leydig细胞的分离纯化技术方法研究

【摘要】 目的 为了深入研究Leydig细胞及其调控因素 ,采用本实验以获取较高纯度 (6 0 %~ 85 % ,甚至 90 %以上 )的Leydig细胞。 方法 利用Percoll不连续密度梯度离心法 ,可以简便迅捷地获取较高纯度 (6 0 %~ 85 % ,甚至 90 %以上 )的Leydig细胞。 结果 得到较高纯度 (6 0 %~ 85 % ,甚至 90 %以上 )的Leydig细胞。 结论 本实验采用简便迅捷的方法 ,虽然不能得到高纯度 (95 %以上 )的Leydig细胞 ,但可以用简单的设备 ,较短的时间 ,获得满足实验要求的较高纯度 (6 0 %~ 85 % ,甚至 90 %以上 )的Leydig细胞 ,并通过体外细胞培养 ,取得具有生物学活性的Leydig细胞克隆 ,为进一步研究这种细胞提供了条件。

【Abstract】 Objective In order to investigate the rat Leydig cells and the regulate factors of this kind of cells.By using the method of purification of rat Leydig cells which was described in this paper,we could obtain highly purer rat Leydig cells (60%~85%,even more than 95%). Methods We could obtain highly purer rat Leydig cells in an easier way by a four-layer discontinuous Percoll gradients(26,34,40,and 60% Percoll in PBS)centrifugation. Results This method of separation of rat Leydig cells by a four-layer discontinuous Percoll gradients centrifugation could isolatate a higher degree of purity (60%~85%,even more than 90%) of rat Leydig cells. Conclusions In contrast with the other methods,although this purification of rat Leydig cells described in this paper could not separate a highest degree of purity(more than 95%) of rat Leydig cells,but this method could obtain a higher purity (60%~85%,even more than 90%)by simple equipments in a shorter time,which satisfied the requirements of the experiments completely, and we also investigate this cells more by using the rat Leydig cells clone which was obtained with culturing the purified rat Leydig cells which possessed all biological charactors in vitro
