紫柏湿疹颗粒抗变态反应及抑制大鼠亲同种细胞抗体介导的肥大细胞脱颗粒实验研究

【摘要】 目的 观察紫柏湿疹颗粒 (ZBSZ)抗变态反应作用。方法 观察紫柏湿疹颗粒对Ⅰ型变态反应的影响 ,采用 4 8h同系大鼠被动皮肤过敏反应 (PCA)方法 ,对Ⅱ型变态反应的影响采用反向皮肤过敏反应 (RCA)法 ,对Ⅲ型变态反应的影响采用Arthus反应 ,对Ⅳ型变态反应的影响采用 2、4二硝基氯苯 (DNCB)法。本实验还观察了紫柏湿疹颗粒对大鼠亲同种细胞抗体介导的肥大细胞脱颗粒及溶血素生成的影响。结果 紫柏湿疹颗粒 2 5g/kg灌胃给药可显著抑制大鼠被动皮肤过敏反应 (P <0 0 5 ) ,并可抑制大鼠亲同种细胞抗体介导的肥大细胞脱颗粒 ,1 5、2 5g/kg抑制率分别为37 8%和 4 1 5 % ;紫柏湿疹颗粒显著抑制大鼠反向皮肤过敏反应 (RCA) ,0 5、1 5、2 5g/kg抑制率分别为 6 5 0 % ,6 8 1% ,和 70 2 % ;能明显抑制大鼠Arthus反应 ,对 2、4二硝基氯苯 (DNCB)所致小鼠迟发型变态反应 (DTH)亦有显著抑制作用 ,并可使小鼠胸腺指数和脾指数明显降低。结论 紫柏湿疹颗粒对Ⅰ~Ⅳ型变态反应及亲同种细胞抗体介导的肥大细胞脱颗粒有显著抑制作用

【Abstract】 Objective To observe the effects of Zibaishizhen Granules (ZG) against allergic reactions. Methods The 48-hour rat-homologous passive cutaneous anaphylaxis (PCA) was used to observe the effects of ZG on type Ⅰ allergic reaction; the reversed cutaneous anaphylaxis (RCA) was used to observe the effects of ZG on type Ⅱ allergic reaction; Arthus reaction was used to observe the effects of ZG on type Ⅲ allergic reaction; 2,4-Dinitrochlorobenzene (DNCB) method was used to observe the effects of ZG on type Ⅳ allergic reaction; and the effects of ZG on degranulation of mastocytes mediated by rat homocytotropic antibodies, and on the production of hemolysin were also observed in the experiment. Results Intragastric administration of ZG at a dose of 2.5 g/kg could markedly inhibit rat PCA (P<0 5); the same administration of ZG at the doses of 1.5 g/kg and 2.5 g/kg could inhibit degranulation of mastocytes mediated by rat homocytotropic antibodies, wit the inhibitory rates as 37.8% and 41.5%, respectively. When being administered at the doses of 0 5 g/kg, 1.5 g/kg and 2.5 g/kg, ZG could inhibit rat RCA, with the inhibitory rates as 65 0%, 68 15 and 70.2%, respectively. ZG could also markedly inhibit rat Arthus reaction and mouse delayed type hypersensitivity induced by DNCB, and decrease mouse thymus index and spleen index. Conclusion ZG can exert significant inhibitory effects on the allergic reactions of type Ⅰ to type Ⅳ, and on degranulation of mastocytes mediated by homocytotropic antibodies.
