【摘要】 目的建立特异、快速、准确的SARS冠状病毒检测方法 ,并探讨其临床应用价值。方法根据WHO公布的SARS冠状病毒的基因序列 ,采用套式RT -PCR方法对 12 7份SARS患者不同病程的血清标本中SARS病毒核酸进行扩增 ,用敏感度 ,特异度 ,阳性预测值 (PPV) ,阴性预测值 (NPV) ,准确度等对该方法诊断SARS的效果进行评价。结果病程在 10d以内的 32份血清标本中 ,13份阳性 ,敏感度 4 0 .6 % ,特异度 10 0 .0 % ,阳性预测值 10 0 .0 % ,阴性预测值 6 6 .1% ,准确度 72 .5 %。结论本研究建立的套式RT -PCR方法可作为发病早期SARS病人的辅助诊断。更多还原

【Abstract】 Objective To develop a sensitive, specific and rapid nested RT-PCR assay for detecting severe acute respiratory syndrome coronavirus (SARS-Cov) and to evaluate its value for clinical application. Methods The primers were designed and synthesized according to the published sequence of SARS-Cov genome. The fragments of SARS-Cov in 127 serum specimens of SARS patients were amplified by nested RT-PCR assay. The sensitivity, specificity, positive predictive value (PPV), negative predictive value (NPV), and accuracy were used to evaluate the significance in diagnosis of SARS. Results Of 32 serum specimens within 10 d-course of the disease, 13 specimens were positive, and the sensitivity, specificity, positive predictive value, negative predictive value, and accuracy were 40.6%, 100.0 %, 100.0%, 66.1%,and 72.5% respectively. Conclusions The nested RT-PCR may be a rapid, accurate and efficient way for assisting early diagnosis of patients with SARS.更多还原
