【摘要】 目的 :探讨尿毒净Ⅱ号对人肾小球系膜细胞CyclinD1、CDK4的影响。方法 :应用流式细胞术、免疫组化、Westernblot方法 ,在培养的人胚胎肾小球系膜细胞 ,分别检测尿毒净Ⅱ号对系膜细胞CyclinD1、CDK4以及DNA倍体的影响。结果 :尿毒净Ⅱ号明显抑制血清加内皮素 - 1(ET - 1)刺激的系膜细胞CyclinD1、CDK4表达 ,增加G0 /G1期细胞 ,降低S +G2 /M期细胞 (与对照组比较P <0 .0 1) ,呈一定的量效依赖关系。结论 :尿毒净Ⅱ号通过抑制细胞CyclinD1、CDK4,阻止系膜细胞由G1期进入S期 ,延缓了细胞周期进程 ,从而抑制系膜细胞增殖。更多还原

【Abstract】 Objective:To investigate the effects of Niaodujing Ⅱ on Cyclin D 1 and CDK 4 in cultured glomerular mesangial cells in human.Methods:Study in vitro, Cyclin D 1 and CDK 4 of cultured mesangial cells (MsC) in human embry, stimulated by sera and ET-1 following incubation with Niaodujing Ⅱ was measured by flow cytomertry (FCM),immunohistochemistry and Western blot method. Results:Niaodujing Ⅱ inhibited the sera and Endothelin-1(ET-1) stimulated expression of Cyclin D 1 and CDK 4 in a dose-dependent manner. It had an increased percentage of cell in G 0/G 1 phase and a reduced percentage of cell in S phase (P<0.01). Conclusion:Niaodujing Ⅱ induced cell-cycle arrest at G 1 phase by inhibiting expression of Cyclin D 1 and CDK 4.It suppressed the proliferation of MsC by inhibiting DNA synthesis.更多还原
