银杏叶对阿尔茨海默病(AD)治疗作用的实验研究

【摘要】 目的 探讨银杏叶对AD的治疗作用。方法  30只SD大鼠随机均分为模型组、对照组和银杏叶治疗组 ,用Morris水迷宫实验检测大鼠学习记忆能力 ,用旷场实验观察大鼠探究活动能力和紧张度 ,用硫磺素 S法和镀银法检测大鼠海马中老年斑和神经原纤维缠结形成情况。结果 模型组大鼠的学习、记忆功能和探究活动与对照组和银杏叶治疗组相比均明显下降 (P <0 .0 5 ) ,银杏叶治疗组与对照组相比无明显差异(P >0 .0 5 )。模型组大鼠海马可见到老年斑沉积和神经原纤维缠结形成 ,银杏叶治疗组仅有少量的老年斑沉积和神经原纤维缠结形成 ,而对照组大鼠未见老年斑沉积和神经原纤维缠结形成。结论 银杏叶提取物可提高大鼠的学习和记忆能力并明显减少老年斑沉积和神经原纤维缠结形成。

【Abstract】 Objective To establish a rat animal m od el of Alzheimer disease and explore the treated effect of gingkgo biloba leaf on AD. Methods Thirty SD rats were randomly divided into m odel group、control group and gingkgo biloba leaf treated group. The learning an d memory functions were estimated by water maze test and the exploratory activit y and tensity were observed by square test. The deposition of senile plaques and the formation of neurofibrillary tangles were observed by thioflavin-S stain a nd silver stain respectively. Results Learning, memory fu nctions and exploratory activity were significantly damaged in the model rats co mpared with control rats and gingkgo biloba leaf treated rats (P<0.05), wit hout difference between control rats and gingkgo biloba leaf treated rats(P >0.05).Neurofibrillary tangles and senile plaques were detected in the hippocam pus in the model rats, less neurofibrillary tangles and senile plaques were dete cted in the gingkgo biloba leaf treated rats, but not detected in the control ra ts.Conclusion The extraction of gingkgo biloba leaf can i mprove rat’s learning and memory functions and decrease the formation of neurofi brillary tangles and the deposition of senile plaques in Alzheimer rat.
