神经节苷脂GM-1对大鼠缺血性脑损伤保护作用的实验研究
Effects of GM-1 on the DNA splitting rate and the expression of Fas and Bcl2 protein in rat following cerebral ischemia reperfusion injury
【摘要】 目的:研究神经节苷脂GM-1对大鼠脑缺血再灌注损伤的保护作用及Fas、Bcl-2表达的影响。方法:建立全脑缺血再灌注模型,应用二苯胺法和免疫组织化学方法观察脑缺血后3h、6h、24h脑组织DNA裂解率变化及Fas、Bcl-2表达,以及GM-1给药后的变化。结果:随着再灌注时间的延长,DNA裂解率变化明显增加,24h达高峰,Fas蛋白表达于再灌注6h达高峰,Bcl-2表达于再灌注3h达高峰,以后逐渐呈下降趋势;GM-1给药组相应时间点的DNA裂解率及Fas蛋白表达明显减少,Bcl-2表达增加。结论:GM-1能抑制脑缺血再灌注后细胞凋亡的发生,对脑缺再灌注损伤具有明显的保护作用。
【Abstract】 Objective To study the neuroprotective effects of ganglioside GM-1and its effects on DNA splitting rate and the expression of Fas and Bcl-2protein.Method In the animal model of global cerebral ischemia reperfusion,using immunohisto -chemistry method,the authors observed the change of DNA splitting rate and the expression of Fas and Bcl-2protein at3h,6h,24h after reperfusion and change after GM-1was administered.Results With the prolonging of reperfusion time,DNA splitting rate in-creased,and peaked at24h after reperfusion.The expression of Fas protein peaked at6h after reperfusion.The expression of Bcl-2protein peaked at3h after reperfusion,and gradually decreased.The damage of GM-1group was less than that in the relative time of NS group,and the expression of Fas reduced,the expression of Bcl-2increased.Conclusion GM-1can inhibit the apoptosis of cere-bral ischemia,so it has neuroprotective effects on cerebral ischemia reperfusion injury.
