百合顶芽离体诱导小鳞茎及开花球培育

Induction of Bulblets from Shoot Apex in Vitro and Cultivation of Flower Bulb in Asiatic Hybrid Lily

【摘要】 以亚洲百合顶芽为外植体,进行了离体诱导小鳞茎及小鳞茎培育开花球研究。结果表明:在BA 1- 2 mg/L+NAA 0.1-0.2 mg/L+MS培养基上,顶芽培养产生丛芽的诱导率高达100%。丛芽在MS+BA 0.5 mg/L+NAA 0.05-0.1 mg/L培养基上不断增殖,并逐渐发育成无根苗,增殖系数为6。激素、蔗糖、光照强 度及低温处理等因素影响小鳞茎诱导,丛芽在MS+IBA 0.5 mg/L+质量分数为4%蔗糖培养基上,光照强度 1.5 klx时,诱导形成小鳞茎效果较好,诱导率100%,平均每株诱导小鳞茎1.5个,小鳞茎直径0.96 cm。小鳞 茎经5℃低温处理2周后,在BA 0.1 mg/L+NAA 0.5 mg/L+MS培养基中诱导出小鳞茎,诱导率82.5%,每块鳞 茎片诱导4.2个小鳞茎,直径0.29 cm。直径0.8 cm以上试管鳞茎,经过5-8℃低温处理4周后,在福建省南 平海拔800 m处栽培8个月,收获的种球中45.8%达到开花球标准。试管鳞茎开花性状稳定。

【Abstract】 In this experiment bud induction from shoot apex, bulblet induction from buds in vitro culture and flower bulb cultivation from bulblet in field were carried out in Asiatic hybrid lily. The results showed that bud induction rate was as high as 100% on the MS medium containing BA 0.5 mg/L and NAA 0.1 - 0.2 mg/L. The small bud proliferated constantly and became bud seedlings, and their proliferation coefficient was six on the MS medium with BA 0.5 mg Land NAA 0.1 - 0.2 mg/L. Bulblets could be induced by bud seedlings in test tubes. Many factors like plant hormone, sugar, illumination intensity and low temperature affected bulblet formation. Under the condition of illumination intensity for 1.5 klx, on the MS added with IBA 0.5 mg/L + sugar 4% , bud seedlings have a better effect on inducing growing bulblets, with 100% induction rate and 0.96 cm diameter of inducing bulblet, and 1.5 average bulblets in one bud seedling. Under the condition of low temperature 5℃ for 2 weeks, the test-tube bulblet regeneration occurred on the MS medium supplemented with BA 0.1 mg/L + NAA 0.5 mg/L, their diving rate reaches 82.5% ,4.2 little bulbs for each bud scale and 0.29 cm diameter of little bulb. By treating with low temperature 5 ~ 8℃ for 4 weeks, the test-tube bulbs, the diameter of which was over 0.8 cm, were cultivated for eight months at a mountain of the elevation of 800 m near Nanping in Fujian. 45.8 % of the new bulbs harvested were big enough and up to the standard of commercial bulbs, available to produce cut flowers.Their ornamental characters were stable.
