硫化物氧化兼性自养金色链霉菌LD48培养的影响因素研究

【摘要】 硫化物氧化、兼性自养金色链霉菌LD48在以Na2S为唯一硫源的条件下对硫化物氧化酶产生的最适接种量为1%。在试验的pH值范围内,起始培养最适pH值为7.4。随着培养液中乙醇体积分数由1%~5%增加,菌体生长和硫化物氧化活性的积累都在下降。酵母提取物是一有效的生长刺激因子,在无机氮源(NH4)2HPO4的基础上添加0.15%的酵母提取物,能够提高菌体生物量95%。S2O32-与SO42-对菌体生长和代谢产物积累都有严重的抑制作用。培养过程分析表明,培养56h硫化物氧化酶产生达到稳定水平。

【Abstract】 The optimal proporation of inoculation of facultative autotrophic Streptomyces aureus LD48,a strain of sulfide-oxidizing was 1% for the formation of sulfide oxidase under the condition of taking the Na2S as the sole sulfur source.The optimal pH was 7.4 in the range of pH experimented.The growth and accumulation of sulfide-oxidizing activity were decreased with the increasing of alcohol concentration from 1%~5%.The yeast extract was an effective growth stimulator and could raise biomass by 95% by adding it on the basis of inorganic nitrogen (NH42HPO4.Both S2O32- and SO42- had a serious inhibition for growth and sulfide oxidase formation.It was shown by analysis of cultural time course that the formation of sulfide oxidase reached to stabilizing level at 56?h during cultivation period.
