ALA-PDT对多种白血病细胞破坏作用的实验研究

【摘要】 目的:本研究主要观察相同条件的5 氨基乙酰丙酸的光动力疗法(ALA PDT)对不同种类的白血病细胞株生存率的影响,以及细胞死亡类型的差异。方法:选择5种白血病细胞(K562、HL60、U937、MOLT 4和6T CEM)进行比较。用MTT法检测细胞的存活率,用AnnexinV FITC PI双染法检测细胞不同死亡类型的比例。结果:不同细胞对相同条件的ALA PDT的敏感程度不同,依次为U937<MOLT 4<6T CEM<K562<HL60,在ALA和光照共同作用下细胞的存活率明显低于单一因素作用引起的细胞存活率。不同细胞的死亡类型不同,其中MOLT 4和U937细胞以早期凋亡为主,K562和HL60细胞以晚期凋亡为主,而6T CEM细胞以坏死为主。结论:ALA PDT能够灭活白血病细胞,不同种类的细胞对其敏感度不同。

【Abstract】 Objective: To investigate the effect of ALA-PDT under the same experimental conditions on the survival rate of various kinds of leukemia cells and the difference in death model. Methods: We selected five kinds of leukemia cell lines to research and the survival rate of the cell was assessed by MTT method, the death model was inspected by Annexin V-FITC/PI. Results: Different kinds of cells have different sensitivities under the same experimental conditions of ALA-PDT. The sensitivity decreased in the following order U937<MOLT-4<6T-CEM<K562<HL60. Cells treated by ALA combined with irradiation had significant lower survival rate than by single factor. Different kind of cell has different death model, early apoptosis is the main death model in U937 and MOLT-4 cells, late apoptosis is the main death model in HL60 and K562 cells, but most 6T-CEM cell presented necrosis. Conclusion: ALA-PDT can kill leukemia cells and the different kinds of cells have different sensitivity to it.
