氧化苦参碱与国产α-lb和α-2b干扰素对大鼠星状细胞增殖和胶原合成的抑制作用
Inhibition of oxymatrine and homemade interferon-α1b and α2b on the proliferation and collagen synthesis of rat hepatic stellate cells
【摘要】 目的 研究氧化苦参碱与国产α lb和α 2b干扰素对大鼠星状细胞增殖和胶原合成的影响 ,为应用氧化苦参碱防治肝纤维化提供理论依据。方法 分离、培养大鼠星状细胞 ,采用MTT法和3 H 脯氨酸掺入法分别测定细胞增殖和胶原合成。结果 氧化苦参碱在 5~ 5 0 0 μg·ml-1浓度范围内、2种α 干扰素在 5 0 0~ 10 0 0 0U·ml-1浓度范围内能呈剂量依赖性抑制星状细胞的增殖和胶原合成 ,提高药物浓度则形成作用平台。结论 氧化苦参碱与α 干扰素在体外均具有抑制大鼠星状细胞增殖和胶原合成的作用 ,这对防治肝纤维化可能有一定的临床应用价值。
【Abstract】 Objective To invesigate the effect of oxymatrine, a Chinese herbal medicine, and interferon-α1b and α2b on proliferation and collagen synthesis of rat stellate cells in vitro. Methods The cell proliferation and collagen synthesis were tested by MTT and 3 H-proline incorporation, respectively. Results The inhibition of proliferation and collagen synthesis by oxymatrine at dose of 5~500 μg·mL -1 , and by interferon-α lb and interferon-α 2b 500~10 000 U·mL -1 , were observed in aconcentration-dependent manner. Conclusions The Chinese herbal medicine, oxymatrine, has the similar inhibiting potential as interferon-α on rat stellate cells in vitro.
