高效液相色谱法测定奥美定中丙烯酰胺的含量
Content determination of acrylamide in polyacrylamide hydrogel by HPLC
【摘要】 目的 建立奥美定中单体丙烯酰胺含量的RP HPLC定量测定方法。方法 以丙烯酰胺标准品为对照进行RP HPLC测定 :AKZONOBELKR10 0 5C18(5 μm ,4 .6× 2 5 0mm)色谱柱 ,柱温 2 5℃ ;流动相由甲醇 水 (2∶3)组成 ,流速为 0 .6ml/min ;进样量为 5 μl;WATERS 2 4 87UV检测器 ,检测波长 2 10nm。 结果 单体分离良好 ,峰面积A与浓度C呈良好的线性关系 ,回归方程为A =4 5 7319345 318.86 0 2C ,相关系数r=0 .9999。加样回收率为 99.8% ,RSD为 0 .4 3%。以保留时间和峰面积作精密度试验 ,RSD分别为 0 .0 34%和 1.4 9%。结论 该方法样品处理简单 ,准确度高 ,精密度好 ,分离效能高 ,适合奥美定中单体丙烯酰胺含量的测定。
【Abstract】 Objective To establish a method of content determination of acrylamide in polyacrylamide hydrogel by HPLC.Methods The chromatographic conditions were as follows: Column: AKZONOBEL KR100-5C18(5μm, 4.6×250mm); Detection wavelengh: 210nm; Mobil Phase: methanol-water (2∶3); Flow rate:0.6ml/min; Detector: Waters 2487UV-VIS detector. The standard sample was used as control.Results Acrylamide was perfectly separated from polyacrylamide hydrogel. The relationship between the concentration of acrylamide in polyacrylamide hydrogel in range of 2.68×10\+\{-7\}~1.34×10\+\{-6\} g/ml and peak area was favorable linear. The regression equation: \%A\%=457319345318.8602 \%C\%,correlation coefficient \%r\%=0.9999.The recovery rate was 99.8%, RSD=0.43%. And in precision test RSD(A)=1.49%,RSD(tR)=0.034%.Conclusion The method is brief, high precise, high separate efficacy, and suited for the content determination of acrylamide in polyacrylamide hydrogel.
