用灭活哈维弧菌菌苗浸泡免疫大黄鱼的研究

【摘要】 用经福尔马林灭活的哈维弧菌 (Vibrioharveyi)菌苗对大黄鱼 (Pseudodciaenacrocea)幼鱼进行浸泡免疫接种 ,分别以 1.0× 10 8cells/mL、1.0× 10 7cells/mL和 1.0× 10 6cells/mL的菌液浓度 ,浸泡处理鱼体 1min、10min和 6 0min后 ,放入设置在象山港内的网箱中饲养 ,并且在浸泡免疫接种后的 12 6d内 ,定期检测试验鱼的免疫保护力 (RPS)。结果表明 ,在浸泡免疫接种后的 5 6~ 77d内 ,受免鱼的RPS达到了 95 %~ 10 0 % ,随后RPS稍有下降 ,但是 ,直到免疫浸泡接种后第 112~ 12 6天时 ,供试鱼的RPS仍达到 70 %~ 80 %。从设置在野外的网箱中供试鱼的统计结果看 ,与对照组相比 ,免疫鱼疾病发生较少 ,发病时期推迟 ,病程缩短 ,成活率提高 (80 .0 %~95 .0 % ) ,明显高于对照组。对不同日龄大黄鱼幼鱼进行浸泡免疫的试验结果表明 ,4 0日龄、75日龄和 10 0日龄的大黄鱼幼鱼均能在免疫接种后获得特异性免疫保护力。在第 1次免疫接种 5 6d后以同样方式加强免疫 ,试验鱼在 112d内保持了很高的RPS水平 ,明显地高于同期的 1次免疫接种鱼体。

【Abstract】 Large yellow croaker(Pseudodciaena crocea)were immunized by immersion with formalin killed Vibrio harveyibacterin with the concentration of 1.0×10 8 cells/mL, 1.0×10 7 cells/mL and 1.0×10 6 cells/mL, and the treating time are 1 min, 10 min and 60 min respectively. After immersion, the fish was cultured in net cages. Relative survival percent (RPS)was monitored 8 to 18 weeks post immersion. It was showed that during 8~11 weeks post immersion, RPS was the highest,and it reached 95.0%~100.0%, and then slowly declined to 70.0%~80.0% during 16~18 weeks. Field statistics revealed a trend that the immunized groups infected with less diseases, diseases broke later, and the fish recovered sooner. The survival percent of the immunized groups reached 80.0%~95.0%, apparently higher than control groups. The fish at different life time, 40 days, 75 days and 100 days post hatch, respectively, were immunized and RPS was tested afterwards. The results indicated that 40 days old juvenile also acquired protective immunity by immersion. Booster immunization was carried out 56 days after the first immersion. High RPS level was detected during 8~16 weeks post boosting,which was much higher than the once treated group.
