钝顶螺旋藻Fe-SOD基因的克隆及序列分析
Cloning and sequence analysis of a superoxide dismutase gene from Spirulina platensis.
【摘要】 参照GenBank中Fe-SOD基因序列,根据SOD的保守区域设计引物,以钝顶螺旋藻(Spirulinaplatensis)基因组DNA为模板,PCR扩增获得大小为528bp的片段.序列分析表明,其与集胞藻(Synechocystissp.PCC6803)、念珠藻(NostoccommuneDRH1)等物种的Fe-SOD基因的同源性分别为75%、71%,且由该序列推导的氨基酸序列与集胞藻、念珠藻等种属的Fe-SOD相比,同源性为68%、67%.表明该片段为钝顶螺旋藻Fe-SOD基因的部分序列.
【Abstract】 Superoxide dismutase (SOD) plays an important role in all aerobic organisms. Fragment of Spirulina platensis SOD gene was cloned and sequenced. The SOD protein is predicted to have 170 animo acids and a molecular mass of 18.7 kD. The ORF sequence of obtained SOD gene from Spirulina platensis shows large homology to Nostoc commune DRH1. The amino acid sequence deduced from the ORF displays homologies of 68%, 67% and 66% to those from Synechocystis sp. PCC 6803, Nostoc punctiforme and Nostoc commune, respectively. It is revealed that the obtained gene fragment indeed encodes an iron-SOD protein.
