【摘要】 目的　探讨乳腺癌MCF 7、肝癌HepG2 和胰腺癌PANC 1细胞系对泰索帝的敏感性及其与Survivin表达水平的关系。方法　台盼蓝拒染法和MTT试验观察MCF 7、HepG2 和PANC 1三种细胞系对泰索帝的敏感性 ;应用RT PCR和免疫荧光方法检测三种细胞系中SurvivinmRNA和蛋白表达。结果　MCF 7、HepG2 和PANC 1三种细胞系均有SurvivinmR NA和蛋白表达 ,阳性率分别为 39.7%、5 8.2 %和 91.4 % ;泰索帝处理后 ,三种细胞的生长曲线呈现递减性抑制 (P <0 .0 1) ;MTT分析显示 ,MCF 7、HepG2 和PANC 1对泰索帝的半数有效量分别为 (14 .4± 1.9)、(31.9± 6 .4 )和 (6 4 .0± 6 .5 )ng/ml。结论　Survivin表达与肿瘤细胞对泰索帝敏感性呈显著负相关 ,Survivin可作为筛选肿瘤化疗药物的新指标更多还原

【Abstract】 Objective To investigate the correlation between chemosensitivity of tumor cells to taxotere and the expression levels of Survivin.Methods Cell survival fractions of MCF-7, HepG 2 and PANC-1 cells treated with taxotere were determined using the trypan blue dye exclusion assay and MTT assay to investigate the sensitivity of all the three cell lines to taxotere. The expression levels of Survivin in the three cell lines were detected by RT-PCR and immunofluorescence analysis.Results All the cells expressed Survivin at mRNA and protein levels. The percentage of Survivin expression in HepG 2, MCF-7 and PANC-1 cells was 39.7%, 58.2% and 91.4% respectively. Cells growth inhibition showed a descending progressive tendency in sequence of MCF-7, HepG 2 and PANC-1 cells treated with taxotere. By MTT assay, the IC50 of taxotere to HepG 2, MCF-7 and PANC-1 cells was ( 14.4± 1.9) ng/ml, ( 31.9± 6.4) ng/ml and ( 64.0± 6.52) ng/ml respectively with the difference being statistically significant.Conclusion The Survivin expression was negatively related to the sensitivity of tumor cells to taxotere.更多还原
