大鼠脑缺血再灌注后半暗带CAD基因的表达改变
Expression of Caspase-activated DNase gene after focal reversible ischemia in rat brain
【摘要】 目的 探讨大鼠局灶性脑缺血再灌注后缺血半暗带Caspase 3激活的DNA酶 (CAD)基因的表达变化规律。方法 线栓法建立大鼠大脑中动脉闭塞 (MCAO)及再通模型。应用RT PCR技术检测MCAO再通后不同时相缺血半暗带皮质CAD基因的表达。结果 脑缺血再灌注 6h ,缺血半暗带皮质CADmRNA显著升高 ,密度比值为 0 .74±0 .0 4 ,再灌注 2 4h达到高峰 ( 1.13± 0 .11) ,再灌注 72h后仍有较高表达 ( 0 .80± 0 .12 )。结论 脑缺血再灌注后CAD基因表达上调 ,可能参与了缺血后神经细胞损伤过程
【Abstract】 Objective To study the regularity of expression of Caspase activated DNase (CAD) gene in ischemic penumbra after rat focal ischemic middle cerebral artery occlusion/reperfusion.Methods The middle cerebral artery of Wistar rats was occluded for 2h by an intraluminal filament and,recirculation was carried out for 6h,12h,24h,48h,and 72h, reperfusion was carried out for 6h,12h,24h,48h and 72h.Reverse transcription polymerase chain reaction (RT PCR) was applied to evaluate the CAD mRNA expression in rat ischemic penumbra tissue.Results CAD mRNA expression was increased since 6h (0.74±0.04) and peaked at 24h (1.13±0.11) after reperfusion in the penumbra,the level still higher at 72h (0.80±0.12).Conclusion These results indicate that CAD gene expression may be subjected to up regulation after reperfusion of cerebral ischemic and play an important role in the course of neuronal injury.
