【摘要】 目的　观察莪术油 (ZTO)在体外对 3种白血病细胞株的抑制作用和刺激人造血祖细胞集落形成的作用。方法　采用细胞培养方法 ,了解ZTO对急性早幼粒细胞白血病系 (HL - 6 0 )、慢性粒细胞白血病系 (K5 6 2 )和急性红白血病系 (U937)细胞株集落残存率的影响和对粒 -单系造血祖细胞 (CFU -GM)、红系造血祖细胞 (CFU -E)和巨核系祖细胞 (CFU -MK)的促增殖作用 ;采用RT -PCR方法 ,分析经ZTO作用后的白血病染色体易位产生的嵌合基因。结果　ZTO对白血病HL - 6 0、K5 6 2、U937细胞株均有不同程度的抑制作用 ,其中对HL - 6 0的抑制作用最强。ZTO对CFU -GM、CFU -E、CFU -MK的集落产率有明显的刺激作用 ,其中对CFU -E的刺激作用最强。净化后 ,白血病HL - 6 0、K5 6 2、U937细胞株的不同表达基因均有不同程度的减弱。结论　ZTO具有抑制白血病细胞生长的作用 ,同时对正常造血细胞有一定的保护作用。更多还原

【Abstract】 OBJECTIVE To observe the inhibiting effect to three kinds of leucocy themia cells and stimulating effect to hermatopoietic cells.METHODS A group of leucocy themia cells HL-60,K562 and U937 were cultured to study the influence of Zedoary turmeric oil to their survival rates.Meanwhile,CUF-GM,CFU-E and CFU-MK cells were cultured to investigate their reproducing abilities.Furthermore,RT-PCR was adopted to analyze the table gene of Leucocy themia chromosome,which product the moving.RESULTS Zedoary turmeric oil can inhibit leucocy themia cells in different degree with the strongest inhibition to HL-60.Zedoary turemetric oil can also stimulating CFU-GM,CFU-E and CFU-MK with the strongest effect on CFU-E.CONCLUSION Zedoary turmeric oil can inhibit the growth of Leucocy themia cells and protect healthy hematopoietic cells.更多还原
