腈水合酶的发酵及酶学性质
Fermentation and characterization of nitrile hydratase
【摘要】 红球菌RhodococcusrhodochrousJ1在含有脲和钴的培养基中发酵产生腈水合酶,可以催化丙烯腈水合为丙烯酰胺。调节培养条件使发酵液的酶活力达到6150u/ml。温度30℃,反应体系呈中性酶活性很高并且稳定。底物浓度,酶浓度,反应时间的正交实验表明,底物丙烯腈是显著因子,最佳浓度7%,超过此浓度酶会失活;1min内,水合反应转化率可达80%。浓度超过15%的丙烯酰胺会抑制酶的活性。
【Abstract】 When grown on a medium containing carbamide and cobalt,Rhodococccusrhodochrous J1 produced nitrile bydratase,an enzyme which hydrolyzes acrylonitril into acrylamide with a maximum activity of 6150 u/ml when medium compositions and cultural conditions were adjusted. Nitrile hydratase had high activity and stability at 30℃ and in neutral pH solution.The influences of the reaction time and the concentration of substrate and enzyme on the enzyme activity were studied by orthogonal experiment.It was shown that the most significant factor was the concentration of substrate acrylonitrile,and the optimal concentration of acrylonitrile was 7%. Nitrile hydratase started to lose activity when the concentratuion of acrylonitrile was higher than 7%.Under the optimal conditions,the transformation efficiecy of the hydrolyzing reaction could reach 80%within 1 minute. Acrylamide of the concentration over 15% exhibited an inhibition effect on the activity of nitrile hydratase.
