非霍奇金淋巴瘤组织中PTEN/MMAC1/TEP1和MAPK的表达及其相互关系

【摘要】 目的:研究非霍奇金淋巴瘤(non-Hodgkinlymphoma,NHL)中PTEN/MMAC1/TEP1和p42/44MAPK的表达及其相互关系。方法:利用免疫组织化学技术检测77例淋巴结非霍奇金淋巴瘤中PTEN蛋白和p42/44MAPK的表达。结果:在NHL中,PTEN蛋白和p42/44MAPK的阳性率分别为91%(70/77)和84.4%(65/77),PTEN蛋白阴性表达率为9.1%(7/77);PTEN蛋白和p42/44MAPK的阳性表达强度之间呈明显负相关(r=-0.6452);低度恶性组PTEN蛋白的阳性表达强度明显高于高度恶性组(P<0.01);而p42/44MAPK则为高度恶性组的阳性表达强度明显高于低度恶性组(P<0.01);B细胞性NHL中PTEN蛋白和p42/44MAPK的阳性表达强度与T细胞性NHL比较无明显差异(P>0.05)。结论:提示在NHL的发生及进展过程中,可能是由于PTEN蛋白表达降低或缺失,不能有效抑制由致瘤因子异常激活的Ras/Raf/MAPK信号转导通路,使淋巴细胞异常增殖、分化和恶性转化。

【Abstract】 Objective:To study the expressions of phosphatase and tensin homolog deleted on chromosome ten ( PTEN ) /mutated in multiple advanced cancer1 ( MMAC 1 ) /TGF-βregulated and epit-helial cell enriched phosphatase ( TEP 1 ) and p42/44mitogen-activated protein kinase ( p42/44 MAPK ) and their relationship in non-Hodgkin lymphoma ( NHL ) . Methods:The expressions of PTEN protein and p42/44 MAPK were detected by imunohistochemical technique in77samples of the NHL. Res ults:In NHL,the positive rates of PTEN protein and p42/44 MAPK were91% ( 70/77 ) and84.4% ( 65/77 ) respectively,the negative rate of PTEN protein was9.1% ( 7/77 ) 鸦the positive strength of PTEN protein was significantly negatively related to that of p42/44 MAP (r =-0.6542 ) .The expression intensity of PTEN protein in low grade group were higher than that in high grade group (P <0.01 ),and the expression intensity of p42/44 MAPK in high grade group were higher than that in low grade group (P <0.01 ) .The expression intensity of PTEN protein and p42/44 MAPK in B-cell NHL were not significantly different in T-cell NHL (P >0.05 ) . Conclus ions:The decrease or deletion of PTEN protein expression may not be able to effectively inhibit the over-activation of Ras/Raf/MAPK signal transduction pathway ,and this will result in abnormal proliferation,differentiation and malignant transformation in genesis of lymphoma.
