三羟异黄酮对在体大鼠股动脉血管床的影响及其机制
Effects of genistein on femoral vascular bed in vive and its mechanism in rats
【摘要】 目的 :研究三羟异黄酮 (genistein ,GST)对大鼠股动脉血管床灌流压的影响及作用机制。方法 :采用股动脉恒流灌注方法。结果 :不同剂量的GST( 0 4,0 8,1 2mg/kg)注射于股动脉灌注环路时 ,可剂量依赖性地降低股动脉的灌流压 ;预先注射蛋白酷氨酸磷酸酶抑制剂正钒酸钠 ( 5 0 μg/kg)或一氧化氮合酶抑制剂L_NAME ,可部分抑制GST( 0 8mg/kg)引起的效应。结论 :GST降低股动脉血管床的血管张力 ,其机制与酷氨酸激酶抑制和NO释放有关。
【Abstract】 Objective: To explore the action of genistein on the perfusion pressure of femoral vascular bed and its mechanism.Methods: By means of constant flow perfusion in femoral artery,perfusion pressure was recorded.Results: Genistein (0.4,0.8,1.2 mg/kg)decreased the perfusion pressure (PP) of femoral vascular bed in a dose-dependent manner.The effect of genistein (0.8 mg/kg) was partially inhibited by pretreatment of either L-NAME,or by sodium orthovanadate (50 μg/kg),a potent inhibitor of protein tyrosine phosphatase.Conclusion: Genistein can decrease the vascular tone in femoral vascular bed with the underlying mechanism that involves tyrosine kinase inhibition and NO release.
