草莓高效离体再生体系的研究
Study on Highly Efficient Regeneration System in Vitro of Strawberry
【摘要】 以丰香和章姬叶片为试材,研究影响不定芽再生的诸多因素,以建立简单、快速、高效的叶片离体再生体系,为草莓的遗传转化研究与应用奠定基础。结果表明,对某一特定的基因型来说,外植体类型和外源激素种类对其再生芽发生频率和数量都有明显的影响。丰香叶片在MS+TDZ2 0mg/L+2,4-D0 1mg/L+IBA0 2mg/L分化培养基上再生频率达83 2%,平均每叶片再生芽数为1 4个;章姬叶片在MS+TDZ4 0mg/L+NAA0 1mg/L+IBA0 2mg/L分化培养基上再生频率达65 7%,平均每叶片再生芽数为1 3个。10~30d叶龄的叶片再生能力较强。另外,8 0mg/LAgNO3能有效地控制外植体的褐化,促进芽的分化。
【Abstract】 Using the leaves of Fengxiang and Zhangji as experimental materials, the factors affecting adventitious shoots regeneration were studied in order to establishment of a reliable, rapid and efficient in vitro regeneration system of leaf and carry out strawberry transformation successfully. The results of this study indicated that frequencies and amounts of shoot organogenesis were significantly affected by types of the explants and hormone concentrations for a given genotype. The regeneration frequency from Fengxiang reached the highest figure of 83.2% in MS+TDZ 4.0 mg/L+NAA 0.1 mg/L+IBA 0.2 mg/L, while Zhangji obtained the highest level of 65.7% in MS+TDZ 4.0 mg/L+NAA 0.1 mg/L+IBA 0.2 mg/L mediums. Number of shoots per leaf disc were 1.4 and 1.3 respectively. The 10~30 days old leaf had higher regeneration ability. AgNO3 played and important role in controlling the browning of explants, and 8.0 mg/L was recommended.
