瓜叶菊的离体培养与快速繁殖

【摘要】 为探索瓜叶菊杂合体母株优良性状的保存方法,以便快速有效地进行瓜叶菊种苗的生产,以瓜叶菊5个不同花色母株的单芽茎段为外植体进行了离体培养与快速繁殖研究.结果表明:1)不同激素组合的培养基对瓜叶菊茎段初始培养中芽诱导的影响因母株花色不同而不同,尤其在芽萌动时间和萌动数量上表现明显,其中以来自母株M1的外植体在含有0.1 mg/L NAA + 2 mg/L BA的A5培养基的诱导率最高,芽萌动早且数目多;2)在继代培养中,在0~2.0 mg/L 范围内,随BA质量浓度的增加,瓜叶菊芽的增殖倍数增大,但当BA为3.0 mg/L时,外植体茎叶随着时间推移逐渐变厚变脆,成愈伤组织状;3)C6培养基(1/2MS+0.5 mg/LIBA+ 2%蔗糖)是瓜叶菊离体生根培养比较合适的培养基,培养到第16天时根系诱导率为100%,平均根数为18条,平均根粗为0.09 cm;来自C6的试管苗移栽成活率可达95%.

【Abstract】 To find out the optimal method to preserve good quality of mother plant with confused genetic backgrounds and to produce seedings of Senecio hybridus effectively and rapidly,in vitro culture and high frequency propagation of Senecio hybridus was studied by means of using stem-segment with single bud as explants.The results show that: 1) There is a genetic difference regarding to effects of different media on bud inducement of stem-segment of Senecio hybridus ,the explants from mother plant M1 cultured on A5 media sprout earlier,and the multiplied rate reached highest;2)When BA concentration range from 0~2.0 mg/L,multiplied rates was ion the rise with the increasing of BA concentration during subculture,but when BA was 3.0 mg/L,stem and leaves of the explant grows thicker and crisper with the passage of time in the condition healed tissue;3)1/2MS+0.5 mg/L IBA+ 2% sucrose was optimal media to induce root,the ratio of root inducement was 100% at the 16 th day,its average number of roots was 18,its thickness is 0.09 cm on average and survival rate of plantlets from C6 media (1/2MS+0.5 mg/L IBA+ 2% sucrose) was 95%.
