地黄饮子含药脑脊液对海马神经元损伤的影响

【摘要】 探讨地黄饮子对β-淀粉样蛋白(Aβ25-35)诱导体外原代培养海马神元损伤的保护作用 把离体培养的海马神经元分为6组:空白对照组、模型组、VE组、中药低剂量组、中药中剂量组和中药高剂量组,分别测定细胞培养液中乳酸脱氢酶(LDH)含量和MTT自动比色微量分析,观察中药地黄饮子脑脊液对受损神经元的影响 表明中药地黄饮子脑脊液能明显降低Aβ25-35诱导的海马神经元细胞培养液中LDH含量,提高细胞生存活力 地黄饮子有效成分能穿透血脑屏障,对抗Aβ25-35诱导的海马神经元损伤,表现出对海马神经元一定的保护作用

【Abstract】 To investigate protective effects of decoction of rehmanniae on cultured hippocampal neurons injury induced by β-amyloid 25-35 protein (Aβ25-35 ). Cultured hippocampal neurons were divided into 6 groups: normal group、model group、vitamin E group、low dose of TCM group、moderate dose of TCM group and high dose of TCM group. Respective measurement of lactic dehydrogenase (LDH) and colorimetric microanalysis of MTT were observed for the effect of decoction of rehmanniae cerebrospinal fluid to injured neurons. High dose of TCM group could significantly decrease the releasing of LDH and increase the live activity of cell. Decoction of Rehmanniae have protective effects on cultured hippocamal neurons injury induced by Aβ25-35.
