大孔树脂吸附和纯化蒲黄总黄酮的工艺研究

【摘要】 以主成分香蒲新苷及异鼠李素-3-O-新橙皮糖苷含量为指标,研究了HZ802非极性大孔树脂吸附和纯化蒲黄总黄酮的工艺条件。结果表明HZ802非极性大孔树脂对蒲黄总酮有良好的吸附纯化性能,其优化的工艺条件为:样品溶液的浓度为50mg/mL(生药量/体积);树脂用量为生药量的1.5倍(质量比);纯化先用2倍树脂柱体积的水,再用2倍树脂柱体积φ=0.2的乙醇;总黄酮的洗脱用2倍树脂柱体积φ=0.5的乙醇;吸附和洗脱流速均为2mL/min。

【Abstract】 An optimum technique for absorbing and purifying the total flavonoids of Pollen typhae with HZ802 non-polarity macro resin was developed, using the contents of typhaneoside and isorhamnetin-3-O-neohesperidoside as the evaluating criteria. The results showed that HZ802 non-polarity macro resin was suitable for absorbing and purifying the total flavonoid of Pollen typhae. The optimum technical operations were as follows:concentration of the sample was 50 mg/mL(weight of the raw material/volume); volume of the resin was 3 times weight of the raw material; purified with distilled water (2 times the volume of the resin), and then, with 20% ethanol (2 times the volume of the resin); 50% ethanol (2 times the volume of the resin) was used to elute the total flavonoids; both the current velocity of absoring and eluting were 2 mL/min.
