同步辐射X荧光分析海州香薷根中铜结合蛋白的微量元素

【摘要】 利用 Sephadex G-50 凝胶过滤从 Cu 处理海州香薷根中分离得到两个不同的铜结合蛋白。用同步辐射 X荧光(SRXRF)分析了两个蛋白的微量元素,结果表明分子量较小的铜结合蛋白 2 与 Cu 络合能力要强于分子量较大的铜结合蛋白 1。另外,Pb、Fe 和 Zn 等金属元素可以与铜结合蛋白竞争结合。两个铜结合蛋白中 S 含量都很低,说明海州香薷根中铜结合蛋白可能并不以含硫量较高的金属硫蛋白(MTs)或植物络合肽(PCs)为主。实验证明 SRXRF 技术可以很好地用于凝胶过滤分离蛋白的元素分析。

【Abstract】 Through the gel filtration chromatography on Sephadex G-50, two different Cu-binding proteins were separated from copper treated Elsholtzia splendens roots. The elements distribution in the two Cu-binding proteins was investigated by SRXRF. The results showed that the Cu-chelate ability of Cu-binding protein 2 is better than that of Cu-binding protein 1. Besides, Pb, Fe, and Zn can compete with Cu to chelate the Cu-binding proteins. The content of element S is very low in the two Cu-binding proteins, which demonstrates that MTs or PCs are not dominant in the Cu-binding proteins in Elsholtzia splendens roots. The experiment also has proved that SRXRF can be used to study elements distribution in the protein separated by gel filtration chromatography.
