盐酸左氧氟沙星壳聚糖药物膜的制备及性能研究
Study on the preparation and characterization of the levofloxacin hydrochloride and chitosan membrane
【摘要】 以盐酸左氧氟沙星(LF)为药物模型,壳聚糖(CS)为载体,制备了药物质量百分含量为0.5%、1.0%、2.0%的药物膜和以壳聚糖为涂层的涂层药物膜.测定了膜的药物的释放性能和力学性能.用红外光谱(FT-IR),X-射线衍射(X-ray)表征了药物膜中各组分的相互作用,扫描电子显微镜(SEM)表征了药物膜的形貌.结果表明:药物膜中LF和CS形成氢键等分子间力,LF吸附在药物膜的表面,而涂层药物膜的表面无明显的LF.随着膜中药物含量增加,药物膜的拉伸强度和弹性模量均下降,分别为29~11MPa,2050~626MPa.涂层药物膜厚度从0.15~0.25mm,拉伸强度和弹性模量为先下降后增加,变化范围为26~18MPa,1257~75MPa.涂层药物膜在pH=5.8的磷酸缓冲溶液中药物释放率达98%的释放时间为6~13h.
【Abstract】 Used levofolxacin hydrochloride (LF) as model drug, chitosan (CS) as delivery, the membrane with different drug contents and the coated membrane which used chitosan as painting lays were prepared. The interaction between LF and CS and the morphology of membrane were investigated by FTIR, X-ray and SEM. The release behavior of LF and the mechanical properties of the membrane were determined. The results showed that the hydrogen bonding between LF and CS was formed in the membrane. LF was absorbed on the surface of the membrane, and little LF could be seen on the surface of coated membrane. While the contents of LF from 0.5?%(wt) to 2.0?%(wt), the tensile strength and the elastic modulus of membrane were decreased from 29?MPa to 11?MPa and from 2?050?MPa to 626?MPa, respectively. The thickness of coated membrane from 0.15?mm to 0.25?mm, the tensile strength and the elastic modulus of membrane were 26~18?MPa,1?257~75?MPa. About 98?%(wt) of LF was released from coated membrane within 6~13?h in the phosphate buffer solution (pH 5.8).
