白细胞介素-2基因佐剂对柯萨奇病毒B3VP1基因疫苗的免疫增强作用
IMMUNE ENHANCING EFFECT OF INTERLEUKIN-2 GENE ADJUVANT ON COXSACKIEVIRUS B3 VP1 DNA VACCINE
【摘要】 目的探讨白细胞介素 2 (interleukin 2 ,IL 2 )基因佐剂对柯萨奇病毒B3(CoxsackievirusB3,CVB3)基因疫苗的免疫增强作用。方法将 pcDNA3/IL 2 +pcDNA3/VP1、pcDNA3/VP1、pcDNA3/IL 2、空白质粒 pcDNA3和生理盐水分别肌内注射免疫BALB/C小鼠 ,1次 /周 ,共注射 3次 ,每次注射后的第 6天断尾取血 ,用微量中和试验方法检测血清中和抗体 ;第 3次注射后的第 7天用 80 0TCID50 CVB3感染小鼠 ,观察各组小鼠的生存情况。结果pcDNA3/IL 2 +pcDNA3/VP1和 pcDNA3/VP1组小鼠能产生中和抗体 ,抗体滴度随免疫次数增加而提高 ,pcDNA3/IL 2 +pcDNA3/VP1组抗体水平高于同期 pcDNA3/VP1组抗体水平 ,但各组小鼠的生存率无显著性差异(P >0 .0 5 )。结论IL 2基因佐剂对 pcDNA3/VP1诱导抗体产生有一定免疫增强作用。
【Abstract】 ObjectiveTo investigate the immune enhancing effect of interleukin-2(IL-2) gene adjuvant on Coxsackievirus B3(CVB3) VP1 DNA vaccine. MethodsBALB/C mice were inoculated intramuscularly three times with pcDNA3/VP1,pcDNA3/IL-2+pcDNA3/VP1,pcDNA3/IL-2,pcDNA3 and saline respectively. Sera were collected on the sixth day after every immunization for measuring neutralizing antibodies, and the number of surviving animals was monitored up to 3 weeks after CVB3 infection. ResultsThe neutralizing antibodies could be induced by either pcDNA3/IL-2+pcDNA3/VP1 or pcDNA3/VP1. The titers of antibody were enhanced as the increase of inoculation times. The antibody titers of pcDNA3/IL-2+pcDNA3/VP1 group were higher than those of pcDNA3/VP1, but the surviving rate of animals showed no difference in the two groups. ConclusionThe immunogenic effect of pcDNA3/VP1 could be enhanced by IL-2 gene adjuvant.
