MTT法在抗病毒药物筛选中的应用
A Simple and Rapid Method of MTT Assay for Evaluation of Antiviral Activity
【摘要】 目的 :采用MTT法评价清热消炎复方制剂体外抗柯萨奇病毒 (CVB3 )、呼吸道合胞病毒 (RSV)效果。方法 :待正常细胞长成单层 ,用病毒感染 ,培养一定时间后 ,根据不同病毒感染细胞出现细胞病变 (CPE)时间的不同 ,加入一定量的MTT液 ,然后测定OD570 值 ,计算病毒抑制率。结果 :MTT法较光镜CPE法更为客观 ,能以OD值变化反映药物对病毒的抑制程度 ,从而获得实验结果 ,且便于统计学处理。结论 :MTT法是一种能广泛应用于抗病毒药物筛选和评价的方法
【Abstract】 Objective: To established a rapid and sensitive method for evaluation of antiviral activity.Methods: The antiviral activity can be assesed as following: After the Hep 2 cells (at a dentisty of 2×10 5 seeded in 40 well tissue culture plates) were infected with CVB 3 and RSV and incubated for 1.5 h at 37 ℃. The virus were removed and different concentration of the test samples were added and incubatded again. When the cytopathic effect (CPE) could be observed at almost every test wells, 50 μl of the MTT solution (5 mg·ml -1 ) was added to each well. After 3 hours’ incubation, the MTT solution was removed and 100 μl DMSO was added to each well. 10 minutes later, the absorbance of the wells was recorded at 570 nm wavelengths for calculating the inhibitory rate of the virus.Results: The MTT assay was more simple, rapid, efficient, acute and objective than the CPE assay.Conclusion: The MTT assay can be widely applied to the screening and evaluation of anti virus substance.
