玉米组织培养及农杆菌遗传转化体系的研究
Study of tissue culture and agrobacterium mediated transformation system in maize
【摘要】 用玉米A188幼胚在2,4-D浓度为2mg·L-1的N6培养基上诱导愈伤组织,出愈率为100%;将愈伤组织转入5mg·L-1 6-BA的N6分化培养基中,分化苗频率在80%以上。当潮霉素浓度为10mg·L-1,可抑制愈伤组织的生长。用农杆菌浸染A188幼胚愈伤组织,GUS瞬时表达检测发现,3.5%的愈伤组织显示蓝斑,说明根癌农杆菌Ti质粒上的T-DNA已经被导入玉米愈伤组织细胞中。
【Abstract】 The young embryo of A188 inbred were cultured on N6 medium to induce callus. When 2,4-D concentration was 2mg/L, the frequency of callus induction reached to 100%. The high frequency (over 80%) was obtained on N6 medium plus 5mg/L 6-BA. The callus of A188 young embryo were highly sensitive to hygromycin. Callus growth could be inhibited completely by 10mg/L hygromycin. The type Ⅱcallus was used as target tissue for transformed with agrobacterium tumefaciens. After infection and coculture, transient expression assay of GUS suggested that 3.5% callus had blue blots.
