魔芋愈伤组织诱导、分化及植株再生的初步研究
Study on Callus Induction, Differentiation and Plant Regeneration of Amorphophallus
【摘要】 采用白魔芋(Amorphophallusalbus)块茎为外植体,以MS培养基为基本培养基,在不同生长调节剂组合的诱导培养基和分化培养基上进行培养。试验结果显示,在MS+0.5mg·L-16 BA+0.5mg·L-1NAA、MS+0.5mg·L-16 BA+0.5mg·L-12,4 D及MS+1.0mg·L-16-BA+1.0mg·L-12,4 D的培养基上容易诱导愈伤组织(诱导效率分别为92%、96%和93%),且愈伤组织容易分化;在分化培养基MS+1.0mg·L-16 BA+0.1mg·L-1NAA及MS+1.0mg·L-16 BA+0.5mg·L-1NAA上,愈伤组织容易分化(分化效率分别为86%和52%)。将在MS+1.0mg·L-16 BA+0.1mg·L-1NAA培养基上分化出的不定根和不定芽转至MS培养基上,30d后培养出完整植株。
【Abstract】 Explants from tuber of Amorphophallus albusTB were cultured in media based on MS medium. Different concentrations of different growth regulators were added into those media. Results showed that callus was induced and differentiated easily from explants cultured in media consisted of MS+0.5 mg·L-16-BA+0.5 mg·L-1 NAA, MS+0.5 mg·L-1 6-BA+0.5 mg·L-1 2,4-D and MS +1.0 mg·L-1 6-BA+1.0 mg·L-1 2,4-D(The induction rates were 92%,96% and 93% respectively); Callus was differentiated easily in media consisted of MS+1.0 mg·L-1 6-BA+0.1 mg·L-1 NAA, MS+1.0 mg·L-1 6-BA+0.5 mg·L-1 NAA(The differentiation rates were 86% and 52% respectively). The adventitious roots and shoots cultured in MS+1.0 mg·L-1 6-BA+0.1 mg·L-1 NAA were transferred into MS medium, 30 days later regenerated plants were cultured from them.
