营养盐对小球藻生长及胞内多糖含量的影响
Effects of Nutritions on the C.vulgaris Growth and on the Content of Saccharide
【摘要】 微藻含有的多糖以及糖蛋白等活性物质具有许多重要的生物活性和生理功能。在24~26℃、自然光照的条件下,首先考察小球藻生长与胞内多糖积累的关系,其次利用正交设计实验考察不同浓度的营养盐对小球藻生长及胞内多糖含量的影响。结果表明,小球藻中多糖的合成与积累主要发生在生长过程的平衡期。利于小球藻生长的营养盐的优化组合为:C6H12O6:2g·L-1,KNO3:0 8g·L-1,NaH2PO4:0 2g·L-1,此时的细胞密度为6 46×107个细胞·L-1培养液。获得最高总糖含量的营养盐的优化组合为:C6H12O6:10g·L-1,KNO3:1 6g·L-1,NaH2PO4:0 4g·L-1。与之对应的总糖含量为:168 1mg·g-1。此结果为进一步从微藻中获取单一组成的、具有重要生物和生理活性的多糖及糖蛋白提供了优化培养微藻的实验依据和规模化生产的基础条件。
【Abstract】 Polysaccharides and glycoprotein in microalgae have many important biological activities and physiological functions. Under the conditions of 24~26℃ and natural light, the relationship between the growth of C.vulgaris and the accumulation of saccharide was investigated, and then the effects of nutritions on the C.vulgaris growth and the content saccharide was studied by mean of orthogonal design test. The results showed that the synthesis and accumulation of saccharide in C.vulgaris occured in the stationary phase. The concentration and optimal combination of nutritions depent on the experimental intention. For the C.vulgaris growth , the optimal combination of nutritions was: C6H12O6: 2 g·L-1, KNO3: 0.8 g·L-1,NaH2PO4: 0.2 g·L-1, the highest cell density was 6.46×107 cells·L-1 culture; and for the content of saccharide, that of nutritions was: C6H12O6: 10 g·L-1, KNO3: 1.6 g·L-1, NaH2PO4: 0.4 g·L-1.Correspond to the results, the content of saccharide was 168.1 mg·g-1 dry weight. The experiment results provided the laboratory foundation and precondition for further optimally culturing C.vulgaris in great scale and further obtaining the important monoforms of protein and saccharide from C.vulgaris.
