一氧化氮对体外培养成骨细胞生长的影响
Effects of NO on growth of osteoblasts in vitro
【摘要】 目的本研究旨在观察不同浓度NO对体外培养兔成骨细胞生长的影响。方法采用组织块细胞培养技术进行兔成骨细胞的分离培养 ,通过碱性磷酸酶 (ALP)细胞化学染色和I型胶原免疫细胞化学染色鉴定成骨细胞。通过测定ALP含量判定NO浓度的改变对细胞生长的影响 ;通过图像分析系统对染色的I型胶原检测 ,了解NO对成骨细胞生长的影响。结果 (1)采用组织块培养技术获得的成骨细胞阳性率达 95 % ;(2 )经AMP法检测上清液中ALP含量 ,不同浓度组Sin 1与对照组比较 ,差异有显著性 (P <0 0 1) ,各组间比较差异也存在显著性 (P <0 0 1)。结论 (1)采用组织块细胞培养技术可获得纯化的成骨细胞。 (2 )NO浓度的增加对成骨细胞生长有明显抑制作用。
【Abstract】 Objective To observe the effect of different concentration NO on growth of osteoblasts in vitro. Methods Osteoblasts were obtained from rabbit costal bone by culture technology and identify by cytochemical staining of alkaline phosphatase (ALP), staining of I collagen protein. The second subculture were exposed to Sin-1(0.1,1.0, 10μmol/L)-NO release dose. The staining imagines of I collages protein were determined by magine analytics instrument after 168 hours. Results 1. Postitive rate of osteoblasts was 95%. 2. Alkaline phosphatase activity in the media collected at 24, 72, 68 hours of culture were determined by Automatic Analyzer, ALP activity in different Sin-1 were statistically different at the same period comparing with control group (P<0.01) ALP activity in different Sin-1 dose 0.1, 1.0, 10μmol/L, were satisfied different (P<0.01).Conclusion Osteoblasts could be obtained by culture technique;Growth of osteoblasts is inhibited by NO concentration rising.
